Embodiment 1

[0039] A method for establishing an EBV virus tree shrew animal model, including animal selection, virus liquid preparation, inoculation, observation of tree shrew status, regular blood sampling after tree shrew inoculation, separation of PBMCs, qPCR determination of EBV copy number in PBMCs, RT-PCR It was established by detecting the expression of EBV-related genes and pathological detection in PBMCs.

[0040] For the animal selection, 13 adult and healthy tree shrews (1-1.5 years old) were selected, male or female, with a body weight of 120-160g.

[0041] The virus liquid preparation is produced by B95-8 culture and lysis, and the cells are purchased from the Shanghai Cell Bank of the Chinese Academy of Sciences.

[0042] Described inoculation, 13 tree shrews are divided into experimental group (n=10) and control group (n=3), represent with Ts1-13, experimental group inoculates 500 μ l EBV virus liquid through nasal cavity nasal drip, is about 3.9 * 10 8 copy; the control g...

Abstract

The invention discloses a method for establishing an EB virus tree shrew animal model. The method for establishing the model comprises the steps of animal selection, preparation of a virus liquid, innoculation, observation of the state of a tree shrew, periodical blood drawing after innoculation for the tree shrew, separation of PBMCs, measuring of EBV copy numbers in PBMCs through qPCR, measuringof expression of relevant EBV genes in PBMCs through RT-PCR, and pathological examination. The method for establishing the EB virus tree shrew animal model is disclosed for the first time, and a basis is laid for better researching EB virus biological characteristics and diseases relevant to the EB virus; the selected tree shrew is a novel experimental animal, and is closer to primates than currently-existing experimental animals; moreover, the tree shrew has the advantages of being susceptible to various viruses, easy to feed, small in size, easy to obtain and the like.

Description

technical field [0001] The invention relates to the field of animal model preparation, in particular to a method for establishing an EBV virus tree shrew animal model. Background technique [0002] Epstein-Barr virus (EBV) was first isolated from African children's Burkitt lymphoma cells by Epstein and Barr in 1964. The infection rate of EBV in the population is as high as 90%, and most of them are in a state of recessive or latent infection. Humans are usually infected in infancy and have no clinical symptoms for life. EBV is transmitted through saliva, and can also be transmitted through blood or sexual channels. Its target cells are B lymphocytes, followed by epithelial cells. Studies have shown that EBV can stimulate the immortalization of B lymphocytes in vitro. EBV is closely related to the occurrence of a variety of malignant tumors, including nasopharyngeal carcinoma, Hodgkin's disease, Burkitt lymphoma, and gastric cancer. The International Cancer Research Center ...
