Abstract

A basic culture medium contains MnSO4.7H2O, ZnSO4.7H2O, Na2MoO4, AlK(SO4)2, H2BO4, CaCl2.2H2O and CoCl2.6H2O; an enriching culture medium contains beef extracts, peptone, NaCl, KH2PO4, agaragar and distilled water, and the pH value is 7.2 to 7.4. The phosphate buffer solution contains NaCl, KCl, Na2HPO4 and KH2PO4, and the pH valueis 7.2 to 7.4. The sludge obtained from a secondary sedimentation tank in a sewage plant is subjected to aerobic culture in the basic culture medium; the cultured sludge is subjected to enrichment culture by the enriching culture medium in a flat plate coating mode, and is put into a biomedical culture box to be cultured for 72h; the enriching culture medium is taken; surface layer sludge is scraped and taken; the phosphate buffer solution is used for repeatedly flushing; then centrifugation is performed by a high-speed centrifugation machine; supernatant fluid is removed; bottom layer sludgeis taken; the sludge is high-concentration halophilic sludge.
