The method comprises the following steps of sampling; and pretreatment and detection for the content of the quinolone in a pretreated target object, wherein the pretreatment includes the steps of mixing a sample with a first extractant to form a first mixed solution, and carrying out ultrasonic treatment for first designated time at a first preset temperature; and cooling the first mixed solution subjected to the ultrasonic treatment to a second preset temperature. Multiple agents are omitted, so the method is more environment-friendly and more healthy to a human body; the time cost is saved, the pretreatment test process is simple, the instrumental analysis time is short and the consumption for a lot of human and material resources is avoided; with a trace test, a detection limit can be lower, and a test instrument is easily available; and an LC-MS / MS analyzer having the high sensitivity and the strong anti-interference capability is used.
