Problems solved by technology

[0015] One of the major drawbacks in solutions for selecting, capturing CTCs by inserting obstacles in the path through which the biological sample flows is caused by the biological and thus morphological changes of the cells themselves subjected to the flow stress.

Embodiment Construction

[0042] Some embodiments may include providing a biological sample comprising a predetermined number of viable cells for analysis. Throughout the specification, "biological sample" shall be construed as a cytological sample of living cells present in various biological fluids (eg, derived from a liquid biopsy and deproteinized in solution). Biological samples are collected from humans and kept in suitable containers and under predetermined environmental conditions until their analysis is performed.

[0043] Prior to analysis, if the biological sample is blood, the biological sample, and thus the living cells therein, can be initially treated by standard protocols (for example, by erythrocyte lysis procedures). Subsequently, the cells can be dispersed in an isotonic saline liquid, (eg, phosphate buffered saline).

[0044] In other cases, such as by way of example and without limitation, the biological sample is not subjected to any preliminary processing in the presence of cere...

Abstract

A method for immobilising live cells within biological samples for analytical and diagnostic purposes that comprises the steps of providing a biological sample containing a predetermined number of cells to analyse; providing a planar support particularly suitable for use in an analytical device, the planar support comprising a surface functionalised with a surface coating comprising a nanostructured material; depositing a laminar layer of said biological sample on said functionalised surface of said planar support in order to cause the cells contained in the biological sample to adhere to thefunctionalised surface of the planar support; and positioning a fixative on the laminar layer of the biological sample. All of the above-mentioned steps are carried out at a temperature below at least25 deg c.

Description

technical field [0001] The present invention relates to the field of instrumental analysis of biological fluids and has particularly been developed with respect to methods for immobilizing biological samples for analytical and diagnostic purposes of liquid biopsy samples. Background technique [0002] Liquid biopsies consist of biological fluid samples taken from various parts of the body (e.g., peripheral blood, bone marrow, cerebrospinal fluid, urine, saliva, sputum, tears, and semen) that contain information relevant to a specific disease or physiological condition of medical significance. cells of potential diagnostic value. [0003] Analysis of cells present in liquid biopsy samples by immunological and molecular characterization is becoming an increasingly important method in diagnostics for screening, prevention, identification and follow-up of pathological conditions, not only for initial diagnosis but also for to assess the efficacy of treatment. [0004] For exam...
