Embodiment 1

[0027] (1) Determination of the initial development site and time point of poplar catkins

[0028] On the campus of Nanjing Forestry University (32° north latitude, 118° west longitude), the female plant of Nanlin 895 fast-growing poplar (NL895), the main species of American black poplar, which has grown for more than 15 years, was selected.

[0029] In March 2018, the female inflorescence florets of NL895 poplar female flowers were collected 1 day after pollination (1DPA), 3 days after pollination (3DPA) and 5 days after pollination (5DPA), placed in FAA fixative solution, and vacuumized. The female florets preserved in the fixative were planed longitudinally, dehydrated with gradient alcohol concentrations, dried, and observed with a scanning electron microscope. The results showed that poplar catkins began to protrude from the epidermal cells of the placenta at the 1DPA time point, and poplar catkin cells elongated rapidly after the 3DPA time point ( figure 1 ). This phys...

Abstract

The invention discloses a regulating gene PdeMIXTA02 for the initial development of poplar catkins of American black poplar and its application, belonging to the technical field of plant genetic engineering. The present invention discloses for the first time the gene PdeMIXTA02 that regulates the initial development of poplar catkins through gene sequence evolution analysis and gene expression analysis at different time points before and after poplar catkins start to develop, and comprehensively verifies the genetic transformation function of Arabidopsis thaliana. This gene belongs to the family of MIXTA transcription factors. Sequence evolution analysis shows that it belongs to the same MIXTA subgroup as the key genes GhMML3 and GhMML4 that regulate the initiation and development of cotton wool. Overexpression in medium can significantly increase the number of epidermal trichomes, and promote the differentiation of Arabidopsis epidermal cell protrusions into epidermal trichomes. The above confirmed that the PdeMIXTA02 gene regulates the initial development of poplar catkins and can be used to breed new varieties of poplar that do not fly catkins.

Description

technical field [0001] The invention belongs to the technical field of plant genetic engineering, and more specifically relates to a regulatory gene PdeMIXTA02 for the initial development of poplar catkins of American black poplar (Poplus deltoides) and an application thereof. Background technique [0002] Poplar (Populus spp.) is the most widely distributed and most adaptable tree species in the world. Poplar cultivation in China is mainly concentrated in North China, Central China, Northeast China and Northwest China. Poplar is not only an important fast-growing and high-yielding timber tree species, but also an important tree species for environmental afforestation; due to its extremely fast growth rate and good cell wall chemical properties, it is also a suitable raw material for the production of second-generation biofuels; its habitat adaptability is strong , and can also be widely used in ecological shelterbelts, three-north shelterbelts, and agricultural and forestr...
