Problems solved by technology

Although QTL-seq conveniently and efficiently narrows down candidate QTL regions, it also has many limitations, for example, it may fail to identify target genes for regions with insufficient meiotic recombination events

Embodiment 1

[0030] 1 source of material

[0031] The present invention uses two pure lines of Porphyra zebra grown in the laboratory, that is, the wild type RZ and the red mutant HT. Spontaneous red mutant HT isolated from wild-type PYL-349. Since each color patch of the offspring four-color patch chimera and the color patches at both ends of the three-color patch chimera have a unified genotype respectively, the four-color patch chimera and the three-color patch chimera are screened out from the F1 leaves (single genotype), construct the CMD population with the above-mentioned method, and evaluate its color trait.

[0032] The test materials are the red pool composed of 24 red individuals and the wild type pool composed of 24 wild type, and the red pool composed of 56 red individuals and 56 The wild type pool consists of two pairs of mixed pools.

Abstract

The invention discloses a method for quickly and accurately positioning chromosomes of laver economic traits and application, and belongs to the field of genomics and molecular breeding. According tothe method, different genotype regions formed by meiosis are distinguished by utilizing the color difference of pigment mutants, and monospores are induced and diffused through asexual reproduction ofa monochromatic block region to construct a filial generation CMD population; the method comprises the following steps: taking a wild parent as a reference genome, performing QTL-seq analysis on an extreme phenotypic pool of a filial generation CMD population by utilizing an SNP / InDel marker, and performing positioning prediction on a quality trait gene or a major QTL by combining KASP and RNA-seq methods. The difficulty in statistical analysis of genetic characters caused by non-uniform genotypes of phylloids of laver filial generations is overcome; the combination method of QTL-seq, KASP and RNA-seq is applied to porphyra character positioning, the thought of porphyra character analysis is greatly expanded, and the method has a good application prospect.

Description

technical field [0001] The invention belongs to the fields of genomics and molecular breeding, and specifically relates to a method and an application for fast and precise positioning of economic traits of laver by using pigment mutants to distinguish meiotic tetramolecules. Background technique [0002] The life history and generational changes of Porphyra are quite different from those of higher plants, which makes the conventional methods of population construction for genetic analysis in higher plants not applicable to Porphyra. Porphyra leaves are developed from conchospores (2n). During the germination of conchospores, meiosis occurs and haploid ordered tetramolecules are produced; it is different from the process of meiosis in animals and higher plants to form a single germ cell, the four cells produced by Porphyra meiosis (with sequence tetramolecule (n)); the genotype of each cell in the tetramolecule can differ due to homologous recombination that occurs during me...
