Abstract

The invention discloses a specific NDRG2 degradation targeting peptide for improving stroke excitatory toxicity injury and application thereof. A specific targeting peptide for rapidly and reversiblydegrading NDRG2 is constructed, wherein design and synthesis of three structural domains are included: (1) synthesizing a cell-penetrating structural domain (CMPD) by utilizing a known membrane-penetrating peptide fragment TAT; (2) specifically binding an NDRG2 structural domain (PBD); and (3) serially connecting a degradation structural domain (CTM / degron). The amino acid sequence of a functionalregion combined with the NDRG2 protein is shown as SEQ. ID. NO.1, the amino acid sequence of the lysosome and proteasome induced degradation peptide fragment is shown as SEQ. ID. NO.2, and the aminoacid sequence of the cell-penetrating peptide TAT is shown as SEQ. ID. NO.3.

Description

technical field [0001] The invention belongs to the technical field of biomedicine, and in particular relates to a specifically degradable NDRG2 targeting peptide for improving excitotoxicity damage of cerebral apoplexy and its application. Background technique [0002] Stroke is the second most fatal and the most disabling disease in human beings, and its prevention and treatment has always been a difficult medical problem in the world. At present, the only proven effective treatment - tPA thrombolytic vascular recanalization therapy only benefits 5% of stroke patients due to the narrow treatment time window (4.5h), which greatly limits its scope of application, and reperfusion injury will also occur after vascular recanalization . In the past 20 years, a large number of gene therapy measures for the pathogenesis of ischemic brain injury (such as excitotoxicity, oxidative stress, inflammation, and apoptosis) reported by a large number of basic studies have failed due to th...
