Abstract

The invention discloses a streptomyces secretory expression vector as well as a construction method and application thereof, and belongs to the technical field of gene engineering. The invention provides a streptomyces secreting type expression vector. The streptomyces secreting type expression vector comprises a sequence as shown in SEQ ID NO. 1. The expression vector is specifically prepared by the following steps: fusing a strong promoter PA4 and a specific secretion signal peptide with the N end of a target gene to be expressed, and inserting into a conventional vector to construct a recombinant plasmid, so that the target gene can be expressed, and the expressed target protein can be secreted outside a cell; in the fermentation culture process of the recombinant strain, the secretory-expressed exogenous target protein can be directly collected from the fermentation liquor. The construction method of the vector is simple and rapid, and the target protein can be directly secreted outside the cell, so that the collection of the target protein is facilitated, and the cytotoxicity caused by heterologous protein accumulation is avoided.
