[0014] 24 male C57BL / 6 mice were randomly divided into 4 groups, namely solvent control group, ARDS model group (LPS injection), PF-06651600 group and PF-06651600 drug intervention ARDS group, 6 mice in each group, PF-06651600 ( 30mg / kg) was administered by intragastric administration, LPS (20mg / kg) was administered by intraperitoneal injection, and the solvent control group was given the same amount of normal saline. The PF-06651600 group and the PF-06651600 drug-intervention ARDS group were intragastrically administered PF-06651600 15 minutes before LPS injection. Eighteen hours later, the serum of the mice in each group was collected by orbital blood collection, and then the mice were sacrificed, and the lungs were dissected and used 10% formalin was fixed, lung injury and neutrophil infiltration were detected by hematoxylin & eosin staining and scoring, and the contents of IL-6, TNF-α and other inflammatory factors in serum were detected by ELISA.

Abstract

The invention provides an application of a Jak3 inhibitor to preparation of a medicine for treating acute respiratory distress syndrome. The chemical name of the Jak3 inhibitor is 1-[(2S, 5R)-2-methyl-5-(7H-pyrrolo [2, 3-d] pyrimidine-4-ylamino)-1-piperidyl]-2-propylene-1-ketone malonate. Animal in-vivo experiment research proves that PF-06651600 has a remarkable treatment effect on the acute respiratory distress syndrome. According to the invention, lung injury and inflammatory change of an acute respiratory distress syndrome mouse model constructed by LPS administration can be effectively reduced, and the Jak3 inhibitor can be used for treatment of the acute respiratory distress syndrome.

Description

technical field [0001] The invention belongs to the field of medicine and relates to a new application of a Jak3 inhibitor, in particular to the application of a Jak3 inhibitor composition in the preparation of medicines for treating acute respiratory distress syndrome, which can alleviate symptoms of acute respiratory distress syndrome. [0002] technical background [0003] Acute respiratory distress syndrome (ARDS) is a common clinical critical illness. Causes include: pneumonia, sepsis, radiotherapy and drug damage, etc. Studies have shown that nearly half of people infected with COVID-19 can progress to ARDS a week after the onset of symptoms; the incidence of ARDS has risen sharply. The fatality rate is as high as 40%, and the treatment window is narrow. The current treatment methods are mainly respiratory support and nutritional support. There is no effective symptomatic treatment drug. The prognosis of patients is poor, the lung function is easily damaged, and the se...
