Problems solved by technology

The infectivity of HCV is weak, there is no ideal cell model, and the HCV virus gene is highly variable, which escapes the recognition of the body's immune system. Therefore, it brings great difficulties to the research of HCV vaccine.
Current treatment methods for HCV infection are limited to interferon and ribavirin with poor efficacy

Embodiment Construction

[0024] The present invention analyzes the amino acid sequences of the antigenic proteins in the NS3 and NS5 regions of HCV, performs epitope prediction analysis, finds multiple single-epitope polypeptide sequences with immunogenic activity, and synthesizes a polypeptide with a length of 10 amino acid residues. Through the immunogenicity test, two polypeptide chains with 10 amino acid residues that can make transgenic mice produce stronger cellular immunity and humoral immunity were screened out. The amino acid sequences of these two polypeptide chains are respectively:

[0025] HCVNS3: pvsylkgssg (10 amino acid residues),

[0026] HCVNS5: nmvyattsrs (10 amino acid residues),

[0027] The purity of the polypeptide vaccine is 95%-99.99%.

[0028] The specific steps of the preparation method of the above-mentioned therapeutic polypeptide hepatitis C vaccine are as follows:

[0029] 1. Solid-phase synthesis of peptides

[0030] In this study, a linear peptide vaccine was synthe...

Abstract

The present invention relates to therapeutic polypeptide hepatitis C vaccine and its preparation, aims at providing efficient method of preventing and treating HCV infection. Through the analysis on the amino acid sequence of HCV NS3 region and NS5 region antigen protein and the predication analysis on antigen epitope, several single epitope polypeptide sequences with immunogen activity are found and corresponding polypeptides in the length of 10 amino acid residues are synthesized by the present inventor. Through immunogenicity test, two polypeptide chains capable of enabling transgenic mouse to generate powerful cellular immunity, in the length of 10 amino acid residues and humoral immunity and in the purity of 95-99.99 % are screened out. These two polypeptide chains are dissolved in medical distilled water to prepare the vaccine solution.

Description

Technical field: [0001] The present invention relates to a therapeutic polypeptide hepatitis C vaccine and its preparation method. Specifically, it is an artificially synthesized polypeptide that can be used to treat hepatitis C virus infection. The polypeptide sequence is selected from the amino acid sequence of hepatitis C virus. It has significant activity of activating humoral immunity and cellular immunity. Background technique: [0002] Hepatitis C virus (HCV) is the main pathogen of post-transfusion hepatitis. More than 70% of acute HCV infection will turn into chronic infection, and a certain proportion will develop into liver cirrhosis and hepatocellular carcinoma as the course of the disease prolongs. At present, there is no specific treatment for many viral diseases. Vaccines are an effective means to prevent viral diseases. At the same time, people are actively researching, hoping to develop therapeutic vaccines for chronic viral infections. The infectivity of H...
