Problems solved by technology

However, this method requires relatively high conditions. For example, the seedlings need to be planted in a greenhouse with better heat preservation and moisture retention conditions, which is conducive to spray inoculation. It takes about 15 days to see the difference in resistance between different varieties

Embodiment 1

[0032] Embodiment 1: (applying the rapid identification of the method of the present invention to the resistance of watermelon variety vine blight)

[0033] A rapid identification method for the resistance of watermelon varieties to vine blight, the method is carried out as follows:

[0034] (1) Plant seedling preparation: select 24 kinds of commonly used cultivars watermelon in production, 10 kinds of muskmelons, seeds are sterilized, sown, grow to 3-5 true leaves after emergence, and set aside;

[0035] (2) Preparation of the spore suspension of the pathogen spores: inoculate the pathogens of the melon spores (from the Research Office of Economic Crop Diseases of the Institute of Plant Protection and Microbiology, Zhejiang Academy of Agricultural Sciences, sampling, isolation and cryopreservation of the diseased plants in the field) On the PDA medium, treat 4 days under 40W ultraviolet lamp (12h ultraviolet lamp / 12h dark) to induce sporulation; wash the spores with sterilize...

Embodiment 2

[0058] Embodiment 2: (applying traditional method to the identification of the resistance of watermelon cultivar to vine blight)

[0059] (1) Plant seedling preparation: select 10 different watermelon varieties of disease index distribution among the embodiment 1, 10 muskmelon varieties (see attached table 2 for specific variety name); Sowing in the same way, when the seedlings grow to 3-5 true leaves, set aside;

[0060] (2) Bacteria bacterium preparation: with embodiment 1;

[0061] (3) preparation of the spore suspension of the pathogen spores: the same as in Example 1;

[0062] (4) spore inoculation: the spore suspension of step (3) is evenly sprayed onto the plant blade with a sprayer, as the criterion without dripping water, 30 seedlings of every kind, and the plant that is not inoculated with pathogenic bacteria is used as a recessive control;

[0063] (5) heat preservation and moisturizing: keep the seedlings in step (4) at a temperature of 25-30°C in a greenhouse or g...

Abstract

The invention discloses a rapid identification method of melon gummy stem blight resistance, belonging to the plant disease resistance identification technical field. The method comprises steps as follows: (1) preparing a plant seedling; (2) preparing gummy stem blight bacteria spore suspension; (3) preparing a culture dish; (4) preparing a leaf; (5) spore inoculation; (6) thermal-insulation and moisturizing cultivation; and (7) investigation of state of disease: disease grade and disease index are investigated 3 to 5 days after inoculation and cultivation. The method has a clear technical basis, and is characterized by simple equipment, convenience and speediness as well as accurate judgment, and the like; compared with traditional identification methods, the method not only can save more than 10 days, but also can simplify the identification and inoculation conditions, lower the cost of identification, and improve the working efficiency. The method can be used in melon pathogenic bacteria pathogenicity determination or melon species resistance identification.

Description

technical field [0001] The invention relates to the technical field of identification of plant disease resistance, in particular to a rapid identification method for resistance to vine blight of melon varieties. Background technique [0002] Watermelon plants are widely cultivated all over the world, and are popular with consumers and farmers for their high nutritional value and good output value. There are many varieties of melons, but the stem blight of melon plants is a very serious disease, and breeders, plant pathologists and farmers are very concerned. Melon blight occurs no matter in the protected area or in the open field. The asexual generation of the pathogen is Deuteromycotina, Didymella bryoniae (Auersw.) Rehn, and the sexual generation is Ascomycotina , melon cocci, the pathogen survives the winter in the diseased residue or soil with pycnidia and ascocases, and in the following year it first infects with conidia, spreads through airflow, water, soil, and can a...
