Embodiment 1

[0029] Embodiment 1: Utilize the method of the present invention to detect 1327 clinical samples

[0030] The cell lines used in this embodiment are as follows: MDCK cells (Shanghai Cell Bank, Chinese Academy of Sciences), LLC-MK 2 (Shanghai Cell Bank, Chinese Academy of Sciences), HEp-2 cells (Shanghai Cell Bank, Chinese Academy of Sciences), RD18S cells (Shanghai Cell Bank, Chinese Academy of Sciences), MRC-5 (ATCC, USA), and other cells are derived from cells cultured and preserved in our laboratory.

[0031] The specimens used in this example came from throat swab specimens collected from adult influenza-like patients in the fever clinics of the First Affiliated Hospital of Guangzhou Medical College and Guangdong Provincial Hospital of Traditional Chinese Medicine from January to September 2009, a total of 1327.

[0032] (1) Cell selection

[0033] The cells are primary or passaged cells, and they are uniform single-layer adherent cells under the microscope. Different ty...

Abstract

The invention provides a fast cultivation and identification method of high throughput virus, which comprises the steps of inoculating cells on a culture plate, inoculating a sample to be cultivated and identified to the cells and then centrifuging, then cultivating, and finally dropping the cell sap to be identified obtained by cultivation to a polyporous glass sheet for immunofluorescence assay. The centrifugal rotation speed is 2500-4000 turns per minute, the time is 30-90 minutes and the temperature is 25-37 DEG C; and the polyporous glass sheet comprises a glass sheet and a film with holes, which are tightly connected. The fast cultivation and identification method of high throughput virus has high detection flux, reduces the sample treatment time and virus cultivation time, increases detection sensibility, has less virus reagent usage, and can be widely applied to virus separation culture and identification of related samples of clinical virus laboratories, public health laboratories and sanitary inspection authorities.

Description

technical field [0001] The invention belongs to the technical field of bioengineering, and in particular relates to a method for rapidly cultivating and identifying high-throughput viruses and its application, which are used for rapidly cultivating and identifying clinical viruses. Background technique [0002] Viral infectious diseases are the main diseases among human infectious diseases today. According to recent data, among the newly discovered infectious diseases in the past 30 years, about 60% of the diseases with identified pathogens are caused by viruses, and about 50% of respiratory diseases are caused by viruses, the typical example of which is SARS coronavirus and highly pathogenic avian influenza virus. Many sudden respiratory infectious diseases, including avian influenza and SARS, usually appear clinically in the form of fever or severe pneumonia, and a definite diagnosis can only be made by relying on etiological examination. The fatality rate of these disea...
