Problems solved by technology

At present, this test can only be done on a fluorescence spectrophotometer. The professional equipment used in the test is not only expensive, but also bulky and cannot be moved. The test results are not continuous, and the online detection of colored soluble organic compounds cannot be realized.

Embodiment Construction

[0034] The present invention will be further described below in conjunction with specific drawings and embodiments. The following embodiments are only used to further illustrate the present invention, and should not be construed as any limitation to the description.

[0035] The invention adopts flow cytometry to continuously detect algae and CDOM on-line. The algae in the detection object contain phycobiliprotein, and its main function is to serve as a light-harvesting pigment complex for photosynthesis, which is formed by the covalent combination of the pigment group phycobilin and the carrier protein. It is divided into four types: phycoerythrin, phycocyanin, phycoerythrin and allophycocyanin. Due to their different contents in different algae, some algae appear red, while others are green. The protein contained in domestic cyanobacteria is mainly phycocyanin. The maximum absorption peak of this protein is around 620nm, which can emit strong fluorescence, has good light abs...

Abstract

The invention relates to a method for detecting alga and colored dissolved organic matters by adopting a flow cytometry. The method comprises the following steps of: (1) alga detection, namely irradiating particles to be detected by a laser beam keeping an angle of 90 degrees with the particles, detecting a forward scattering light (FWC), a side scattering light (SWC) and multi-color fluorescent light of the particles by a detection system, and automatically analyzing by utilizing software, so as to obtain related information such as the fluorescent signal strength of an alga cell, the size of the alga cell and the quantity of the alga cells in unit volume; and (2) detection on colored dissolved organic matters, namely filtering the detected alga particles, irradiating filtrate by a laser beam with the wavelength of 355nm, collecting a light absorption signal and a fluorescent signal of the filtrate in a special wavelength range by the detection system, and carrying out analysis on the colored dissolved organic matters by an intelligent instrument. The method provided by the invention is simple and rapid, the alga and the colored dissolved organic matters (CDOM) in water can be simultaneously continuously detected online, and early warning detection on eutrophication of a water body can be realized.

Description

technical field [0001] The invention belongs to the technical field of online detection and analysis of water quality components, and relates to a detection method for algae and colored soluble organic matter (CDOM) in water, specifically a method for detecting algae and colored soluble organic matter by flow cytometry . Background technique [0002] Algae refers to a class of eukaryotes in the Protist kingdom (some are also prokaryotes, such as algae in the cyanobacteria). Mainly aquatic, without vascular bundles, capable of photosynthesis. It contains phycobiliprotein, whose main function is to serve as a light-harvesting pigment complex for photosynthesis, which is formed by covalently combining the pigment group phycobililin and the carrier protein. It is divided into four types: phycoerythrin, phycocyanin, phycoerythrin and allophycocyanin. Due to their different contents in different algae, some algae appear red, while others are green. The protein contained in dome...
