Abstract

The invention discloses a method of preparing a recombinant human relaxin wild type and a mutant by using a genetic engineering means. The method comprises the following steps of: designing and synthesizing an amino acid codon gene sequence favored by yeast of the recombinant human relaxin wild type and the mutant; (2) designing an amino acid sequence of a human relaxin precursor of the recombinant wild type and the mutant; (3) constructing a secretion type recombinant yeast expression vector of the human relaxin wild type and the mutant; and (4) performing secretion expression of the recombinant wild type and the relaxin mutant. The invention discloses a method of expressing the human relaxin wild type and the mutant by using a genetic engineering means. According to the method, high-efficiency expression of the human relaxin mutant is realized through a pichia pastoris expression system by using a modified secretion type expression vector pPICZ. The method disclosed by the invention has the advantages of high yield, simplicity, convenience, easiness, low cost and the like. The recombinant human relaxin wild type and the mutant have effects of expanding blood vessels and can be used for treating cardiovascular diseases such as hypertension and heart failure, wound healing and treating expanded cervix caused by delivery.
