The method comprises the following steps of: measuring by using a high performance liquid chromatography; taking an appropriate quantity of semen cassiae formula granules; adding diluted hydrochloric acid of 80-120 times in quantity; putting in water bath of 60-100 DEG C; heating and hydrolyzing for 1-2 hours; taking out and cooling; transferring the hydrolysis liquid into a separating funnel; adding an equal quantity of dichloromethane to be extracted so as to separate a dichloromethane layer; repeating for 3-5 times; combining the dichloromethane extract liquor; evaporating in low temperature; dissolving the residue by using a dichloromethane and methyl mixed solution in a volume ratio of 1:1; determining the volume, filtering and taking the subsequent filtrate; respectively absorbing 10 micro-liters of reference substance solutions and test solutions precisely; measuring in accordance with chromatographic conditions; and calculating in accordance with an external standard method so as to obtain the chrysophanol content.
