The method comprises the following steps of: adding to-be-determined Botryococcus liquid in a centrifuge tube, adding particle type xanthomonas polysaccharide, then, placing the centrifuge tube in water bath, and preparing Botryococcus colony formed by dozens even hundreds of cells into aggregation of uniformly distributed single cells or several cells by using ultrasonic waves, so as to meet the requirement of the fluorescent spectrometry on the uniformity of the determined Botryococcus liquid.
