[0037] 1. Materials

[0038] 1) Kelp ( S. japonica ) female and male gametophytes were taken from our laboratory. At a temperature of 17°C and a light intensity of 40 μmol·m -2 ·s -1 , light / dark ratio of 16h / 8h under the conditions of cultivation, the medium is PES, the specific formula see literature: Starr RC, Zeikus JA (1993) UTEX-The culture collection of algae at the University of Texas at Austin. J Phycol 29 (Suppl): 1-106.

[0039] 2) Chlamydomonas reinhardtii ( C. reinhardtii ) is the cell wall deletion mutant strain CC-3491, which was purchased from the Chlamydomonas Center of Duke University, USA. At a temperature of 25°C and a light intensity of 70 μmol·m -2 ·s -1, Light / dark ratio is cultivated under the condition of 12h / 12h, and culture medium is TAP, and specific formula sees literature: Harris, E.H. (1989): The Chlamydomonas sourcebook: a comprehensive guide to biology and laboratory use. Academic Press, San Diego, 780pp.

Abstract

The invention relates to a kelp gametophyte lhcf 6 gene promoter and application thereof, specifically to the sequence of the kelp gametophyte lhcf 6 gene promoter, application of the promoter sequence in promotion of expression of a target gene by algae, a recombinant vector containing the promoter sequence and a preparation method for transgenic algae based on the promoter sequence. According to the invention, the kelp gametophyte lhcf 6 gene promoter obtained through cloning is connected with a fusion expression vector, and an electroporation process is adopted for conversion of Chlamydomonas reinhardtii; it is proved that the promoter has almost similar efficiency in driving expression of a target gene compared with a (CaMV)35S promoter, a basis is laid for further analysis of differential expression mechanisms of the lhcf gene in female gametophyte and male gametophyte, conditions are provided for artificial regulation and control of expression of related genes of algae, and the promoter is originated from kelp, having good safety.

Description

technical field [0001] The present invention relates to the technical field of genetic engineering, in particular, relates to kelp gametophyte lhcf 6 Gene promoters and their applications. Background technique [0002] Light-harvesting protein complex (LHC) is a type of protein embedded in the thylakoid membrane of algae and higher plants to bind antennae or auxiliary pigments and capture and transmit light energy for the photosynthetic reaction center. Under the appropriate light intensity, the LHC of algae plays the role of binding pigments and transfers the light energy captured by the pigments to the photoreaction center for photosynthesis; when the light intensity exceeds the maximum luminous flux required for photosynthesis, the algal cells adjust The expression of LHC gene can reduce the pigment bound on the thylakoid membrane and reduce the transmission of light energy to protect chloroplasts from irreparable damage caused by light inhibition; but under stress condi...
