Problems solved by technology

Each labeling technology has its advantages and disadvantages. For example, isotope labeling technology has high sensitivity and is suitable for long-term tracing, but it has certain radioactive pollution, and the detection is complicated and requires a specific testing site; magnetic labeling technology has a low false positive rate and is non-invasive. However, its detection requires specific instruments and NMR coils of appropriate size; bioluminescent labeling technology, such as pre-transfection labeling methods, is complicated, and the signal intensity in deep tissues is not ideal
[0004] The specific tracer method is to separate lymphocytes from peripheral blood, label them, and then reinfuse them into the body. However, due to the large blood volume of primates and other large animals, reinfusion into the body will cause the target cells to be too diluted and difficult to monitor. Suitable for detection of immune status

Embodiment 1

[0030] see figure 1 The CFSE reagent used in this example was purchased from Molecularprobes Company of the United States, and the red blood cell lysate and mononuclear lymphocyte separation solution (dextran-diatrizoate meglumine) used were purchased from BD Company of the United States.

[0031] The experimental animals used in this example are healthy, clean-grade, male cynomolgus monkeys with a body weight of 7.0Kg, and the experimental animal quality certificate number: SCXK (Guangdong) 2009-0010. The method of animal disposal during the experiment complied with the requirements of animal ethics.

[0032] In order to better highlight the advantages of the present invention, this embodiment is divided into a vascular channel group (group A) and a lymphatic channel group (group B) according to different ways of collecting and infusing mononuclear lymphocytes. The vascular access group (group A) will extract and isolate mononuclear lymphocytes from peripheral blood, label t...

Abstract

The invention discloses an in-vivo tracing method of monocular lymphocytes. The in-vivo tracing method comprises the following steps: collecting lymph nodes in an armpit of a cynomolgus monkey; obtaining the monocular lymphocytes in the lymph nodes by using density gradient centrifugation; carrying out fluorescent staining on the monocular lymphocytes; infusing the stained monocular lymphocytes into connective tissues below thenar eminences of a palmaris of the cynomolgus monkey; collecting the lymph nodes in an armpit of the cynomolgus monkey which is at the same side with the infusion location once again, and detecting the lymph nodes collected again by using a flow cytometer. According to the in-vivo tracing method, the monocular lymphocytes subjected to fluorescent staining can be enriched in the lymph nodes so as to facilitate the immune tracing.

Description

technical field [0001] The invention relates to the field of immune tracing, in particular to a method for tracing mononuclear lymphocytes in vivo. Background technique [0002] Mononuclear lymphocytes play an important role in the body's immune response. At present, the commonly used clinical detection method for immune status is lymphocyte-mediated cytotoxicity test. This method is an in vitro experiment and cannot truly simulate the immune environment in vivo. Due to the complexity of the immune response, the existing immunological detection methods have their limitations. [0003] The existing cell tracking technologies include isotope labeling technology, magnetic labeling technology and bioluminescent labeling technology. Each labeling technology has its advantages and disadvantages. For example, isotope labeling technology has high sensitivity and is suitable for long-term tracing, but it has certain radioactive pollution, and the detection is complicated and require...
