Problems solved by technology

At present, the treatment of patients with chronic n-hexane poisoning includes breaking away from contact, ensuring that patients have adequate nutrition, giving B vitamins, energy mixtures, promoting blood circulation and removing blood stasis, Chinese medicine for dredging collaterals and tonifying kidneys, and supplemented by acupuncture, physical therapy, and limb exercise. There is no specific drug treatment

Embodiment Construction

[0018] First, the isolation and culture of bone marrow mesenchymal stem cells (MSCs):

[0019] Rats were killed by dislocation, sterilized by immersion in 75% ethanol, aseptically peeled off the femur and tibia, removed the metaphysis, exposed the bone marrow cavity, rinsed repeatedly with PBS, collected the cells through a sieve and centrifuged, and weighed in DMEM culture solution containing 10% serum. Cells were suspended and seeded in culture dishes at 37 °C, CO 2 Culture in a saturated humidity incubator, and change the medium after 24 hours. After that, the medium was changed once every 3 days, and the cells were subcultured when the cells reached 80%-90% confluency, and the MSCs were gradually purified by the differential attachment method. Cells with good growth status in the third generation were collected, and flow cytometric detection was performed using monoclonal antibodies CD90 and CD45 to identify MSCs.

Abstract

The invention relates to a particular application of mesenchymal stem cells and solves the problem that in the prior art, no medicines for treating normal hexane-induced peripheral neuropathy are available. The invention provides a novel concept and the particular application for treating the neuropathy. By taking mice with normal hexane-induced peripheral neuropathy as experimental subjects and gait score, nerve electrophysiology (nerve conduction incubation period and nerve conduction velocity), solochrome cyanine dyeing of spinal cord and ischiadic nerve, a scanning electron microscope, myelin basic proteins and low molecular weight neurofilaments as detection indexes, the result shows that the mesenchymal stem cells as a medicine have a remarkable recovery effect on mice with normal hexane-induced peripheral neuropathy and play an important role in treatment of occupational poisoning-induced peripheral neuropathy.

Description

technical field [0001] The invention relates to an application of bone marrow mesenchymal stem cells, in particular to an application of bone marrow mesenchymal stem cells as a medicine for treating peripheral neuropathy induced by n-hexane. Background technique [0002] The nervous system damage in patients with chronic n-hexane poisoning (n-hexane-induced peripheral neuropathy) is caused by the final metabolite of n-hexane and the final poison 2, 5-hexanedione (HD). Its pathological features are axonal neurofilament accumulation and segmental swelling, followed by segmental demyelination, especially long and thick fibers. Many scholars have studied the pathogenesis of n-hexane toxic peripheral neuropathy from the perspectives of axonal skeleton proteins, energy metabolism disorders, and NGF signal transduction pathways, but so far it has not been fully understood. At present, the treatment of patients with chronic n-hexane poisoning includes breaking away from contact, en...
