The preparation method includes following steps: enabling raw cowhide to be expanded in cold water for 2-5 hours and dissolving the raw cowhide in hot water at 200-600 DEG C; processing the dissolved raw cowhide with stirring to prepare a sizing material with a concentration being 3-12%; adding auxiliary materials comprising glycerin accounting for 15-30% by weight of the raw cowhide, zinc sulfate accounting for 0.5-1.5% by weight of the raw cowhide and sodium hydrosulfite accounting for 0.05-0.15% by weight of the raw cowhide with stirring; maintaining the temperature to be 60-75 DEG C after stirring finished and meanwhile adding sodium stearate accounting for 2-4% by weight of the raw cowhide and sodium chloride accounting for 0.06-0.12% by weight of the raw cowhide with stirring; adding oxidized starch accounting for 12-20% by weight of the raw cowhide and the sodium stearate accounting for 0.25-1.0% by weight of the raw cowhide with stirring to perform a reaction for 0.5-2 hours; and performing freeze-drying to obtain the animal glue surface sizing agent. The animal glue surface sizing agent is unique in formula, is simple in a sizing technology process, is low in cost and can significantly enhance surface strength and inherent physical strength performances of finished paper.
