Problems solved by technology

The identification method of emergence rate at the germination stage is mainly aimed at cultivated cotton seeds, while the seed shell of wild cotton is extremely dense and hard, and natural conditions cannot germinate normally, so the traditional low temperature resistance identification method at the germination stage is not suitable for wild cotton seeds; the field identification method at the seedling stage , is greatly affected by environmental factors such as year, region, temperature, humidity, etc., and the repeatability of the experiment is poor
Moreover, there is a lack of a clear evaluation system, and the results are greatly affected by individual subjective factors.

Embodiment 1

[0025] Embodiment 1 adopts the wild cotton seedling stage low temperature tolerance identification method to the tolerance of wild cotton (Thurber's cotton, Krautsky cotton, quasi-like cotton, three-cracked cotton, Raymond's cotton) and cultivated cotton (middle 50) Low temperature levels are identified and evaluated.

[0026] 1. Cotton material:

[0027] Wild cotton (Thurber's cotton, Krautsky's cotton, quasi-like cotton, trifid cotton, Raymond's cotton) and upland cotton (Middle 50) were used as reference materials for cultivated cotton. These wild cottons have been reported to have good low temperature resistance.

[0028] 2. Pretreatment of seeds:

[0029] All the seeds with short fluff are delinted with sulfuric acid, and the complete mature seeds with the same size, plump black and no pests are selected. Cut the chalazal end (blunt end) of the wild cotton seed with a single-sided blade to expose the seed kernel.

[0030] 3. Germination:

[0031] Using the sand bed c...

Abstract

The invention belongs to plant physiology and particularly relates to a method for identifying low-temperature resistance of wild cotton in a seedling stage. The method comprises the following steps: 1) sulfuric acid delinting and incision treatment on seeds; 2) germination accelerating treatment; 3) seedling culture: common seedlings in a 3-5-leaf stage with consistent growth vigor are selected; 4) low-temperature stress treatment: low-temperature stress treatment closer to natural conditions is selected; 5) low-temperature resistance level classification: grading is performed according to damaged conditions of cotton seedlings, and resistance levels of the wild cotton are classified. According to the method, severe low-temperature stress treatment is performed in the seedling stage when the wild cotton is most sensitive to low temperature, so that a low-temperature-resistant cotton material truly with higher resistance level can be screened out; grading is performed according to damaged conditions of cotton leaves, objective classification is performed, and errors caused by human factors are reduced greatly. The method is not limited by seasons, can be used for identifying mass materials and is easy to popularize and use.

Description

technical field [0001] The invention relates to plant physiology and crop cultivation, in particular, the invention relates to a method for identifying low temperature tolerance of wild cotton seedlings. Background technique [0002] The genus Gossypium includes 4 cultivated cotton species and 47 wild cotton species. The cultivated cotton species have undergone long-term artificial directional selection (favored to fiber), and lack the excellent characteristics of adversity survival. However, wild cotton species live in the natural environment for a long time without being subjected to artificial selection pressure, and have excellent traits such as rich diversity and strong stress resistance. To make full use of the unique stress resistance traits of wild cotton resources, it is necessary to identify and evaluate the low temperature tolerance traits of wild cotton resources. The first condition is to find a reasonable and effective low temperature tolerance identification m...
