Abstract

The invention discloses a 60Co gamma ray-induced mutation method for Capsule Epidendrum secundum. The method comprises the following steps: selecting seeds of Epidendrum secundum, determining mediums and culture conditions, dividing the seeds into different groups, subjecting the groups to radiation by 60Co gamma rays, determining a half lethal dose LD50 through statistical analysis and carrying out fitting with related software so as to obtain a linear equation; inoculating differentiation mediums with protocorms germinated from the seeds, carrying out culture and differentiation so as to obtain regenerated plants, investigating the differentiation conditions and morphological indexes of the regenerated plants in 120 d, and analyzing difference between radiation groups receiving different dosages of radiation and a control group; and carrying out mutagenization with 60Co gamma rays under tissue culture conditions, observing mutagenization effect of different dosages of radiation on the seeds of Epidendrum secundum and determining a proper radiation dosage. According to the invention, 60Co gamma ray-induced mutation breeding technology for Capsule Epidendrum secundum is established, provides basis for selection and determination of the proper radiation dosage for radiation-induced mutation breeding of Capsule Epidendrum secundum and enables a novel variety to be bred. The method provided by the invention uses easily available raw materials, can reduce a contamination rate and saves time and cost.
