Abstract

The invention discloses a method for inducing generation of Rosa sterilis tetraploid through a mixed culture method. The method comprises the following steps: Step 1, material preparation: aseptic seedling stem is cut into sections of 0.8-3 cm length with each section at least having a leaf axil; Step 2, preculture: preculture is carried out in a medium MS+0.5 mg / L 6-BA for 0-3 d; Step 3, colchicines light culture: precultured stem sections are transferred into MS+0.5 mg / L 6-BA mediums with 0-200 mg / L of colchicines; Step 4, culture without colchicines: the stem sections are transferred into MS+0.5 mg / L 6-BA mediums without colchicines to be continuously cultured after 7 d of culture in the Step 3; and Step 5, homogenization treatment of variation buds: after 40 d of culture in the Step 4, cultured buds-seedlings with obvious variation is repeatedly cultured in the medium MS+0.15 mg / L NAA+0.5 mg / L 6-BA for five times at 40 d interval. By the induction method, a tetraploid Rosa sterilis new germplasm is obtained. Nutrient components such as Vc, VE and SOD, etc. of Rosa sterilis fruits can be increased as well as fruit width and edible parts.
