Problems solved by technology

[0004] In the past, the prevention and treatment of pear tree rot mainly relied on thiram and other chemicals, but thiram has been banned from being used on fruit trees
At the same time, chemical fungicides are limited due to the tendency of bacteria to develop drug resistance and consumers' increasing concerns about human health, environmental pollution and chemical residues in agricultural products. Therefore, it is urgent to find a new, safe and non-toxic method or means Alternatives to chemical fungicides to control pear tree rot

Embodiment 1

[0036] Example 1 Screening and identification of endophytic fungus Y28

[0037] 1. Isolation, purification and screening of endophytic fungus Y28

[0038] 1.1 Separation of endophytic fungi from leaves and branches (50 samples each)

[0039] The collected samples (the samples come from pear varieties such as the fruit tree practice base of Hefei High-tech Agricultural Garden, Hefei, Hefei, Anhui Province) are washed with running tap water to remove surface impurities, and then surfaced in an ultra-clean workbench Disinfection: Rinse the surface with sterile water, absorb the water with sterile filter paper, soak the tissue in 15% sodium hypochlorite solution for 5-10s and then rinse with sterile water 3 times. The sterilized tissue was cut into the plant tissue with a scalpel, and the internal tissue was cut into small squares of about 5mm×5mm, placed on a sterile PDA, cultured and numbered.

[0040] 1.2 Purification of endophytic fungi

Embodiment 2

[0065] Example 2 Application of strain Y28 in preventing and treating pear tree rot

[0066] Collect the ascospores of strain Y28 and use sterile water to prepare a concentration of 10 8 For the spore suspension of CFU / mL, apply the prepared spore suspension to the completely scraped rot wound, the scraping area exceeds 0.5cm-1.0cm of the lesion, and then apply the spore suspension. The results showed that the rot disease scars coated with the spore suspension healed well, and the diseased tissues were significantly repaired ( Figure 4 ).

[0067] It can be seen that the strain Y28 has the characteristics of safe and efficient prevention and control of rot disease, and this strain is an excellent strain for potential prevention and control of rot disease.

Abstract

The invention provides an endophytic fungus-Talaromyces flavus Y28 with a biocontrol effect. The Talaromyces flavus Y28 is separated from a high-rot-resistance pear tree variety, and a preservation number of the Talaromyces flavus Y28 is CGMCC No.11918. The invention further provides an application of the Talaromyces flavus Y28 to prevention and control of a fruit tree rotten disease. As proved by tests, the antibacterial rate of the Talaromyces flavus Y28 in fungus in-vitro opposing antibacterial tests can be up to 70.50 percent, and the antibacterial rate in annual branch in-vitro secondary screening tests is 55.54 percent, so that the Talaromyces flavus Y28 is a good biocontrol strain for preventing and controlling the pear tree rotten disease.

Description

Technical field [0001] The invention relates to the field of microbiology and biological control technology, in particular, to Demodex yellow Y28 and its application in preventing and controlling fruit tree rot. Background technique [0002] Pear is the third largest fruit tree species in my country. The healthy development of the pear industry not only plays an important role in my country's agricultural production and farmers' income increase, but also has a decisive influence on the world's pear fruit production. [0003] The pathogen of pear tree rot disease is the pathogen of pear tree rot, which belongs to the subphylum Ascomycota fungi, and the morphology of the pathogen is similar to that of the apple tree rot pathogen. Pear rot disease occurs in all major pear-producing areas in my country, especially in Xinjiang, Northwest, and North China. According to the survey, the diseased plant rate of Korla fragrant pear in Xinjiang is 50%-80%, and the diseased plant rate of pear ...
