Embodiment 1

[0050] Embodiment 1, the acquisition and identification of NF-YC9 transgenic plants

[0051] The NF-YC9 gene involved in this embodiment is derived from Arabidopsis thaliana, and its sequence in the Arabidopsis genome is shown in sequence 1 in the sequence table, and sequence 1 consists of 1795 nucleotides; The cDNA sequence of the NF-YC9 gene is shown in sequence 2 in the sequence listing, and sequence 2 is made up of 1613 nucleotides, wherein the 602-1297th position is a coding sequence (CDS); both sequence 1 and sequence 2 are encoded in the sequence listing The protein shown in sequence 3, sequence 3 consists of 231 amino acid residues.

[0052] 1. Identification of NF-YC9 knockout mutants

[0053] NF-YC9 gene knockout mutant T-DNA insertion mutant named nf-yc9-1 was purchased from Arabidopsis Biological Resource Center (ABRC), http: / / www.arabidopsis.org / ), the background is Arabidopsis wild type (Col-0 ecotype). Homozygous was identified by PCR technique.

Embodiment 2

[0079] Embodiment 2, NF-YC9 transgenic plant is to the ABA tolerance analysis test

[0080] ABA can inhibit the growth of plant seedlings. With the increase of exogenous ABA concentration, the inhibition of Arabidopsis wild type (Col-0 ecotype) was enhanced, and the growth of its main root and leaves would be affected to varying degrees. The experiment was repeated three times.

[0081]Arabidopsis wild type (Col-0 ecotype), NF-YC9 gene knockout mutant nf-yc9-1 and NF-YC9 overexpression lines OE1 and OE6 were used as experimental materials. The seeds of each experimental material were sown on MS medium containing different concentrations of ABA (0 μM, 0.4 μM and 0.6 μM) (80-100 seeds were sown for each experimental material). After low-temperature stratification at 4°C for 3 days, move them into the light incubator, observe the growth of Arabidopsis seedlings and measure the root length after 10 days

Abstract

The invention discloses application of NF-YC9 protein in regulating and controlling ABA tolerance of plants. The application specifically refers to application of the NF-YC9 protein or coding gene thereof in regulating and controlling ABA tolerance of the plants. An amino acid sequence of the NF-YC9 protein can be shown as a sequence 3. Experiments prove that tolerance of the plants to ABA is lowered after overexpression of the NF-YC9 protein. Therefore, the NF-YC9 can be used as a growth studying model for studying important genes in regulating and controlling crop growth and development and environment stress and is conducive to deeply clarifying needed molecular mechanism of positive-negative adjusting factors in regulating and controlling response level of the plants to ABA signals, plant growth and stress resistance to meet needs of agricultural production, so that theoretical support is provided for actual agricultural production. The NF-YC9 protein meets development needs of sustainable agriculture and has important practical value and market prospect in the aspects of studying ABA-tolerant molecular mechanism of the plants and crop improvement hereditary characteristics.

Description

technical field [0001] The invention belongs to the field of biotechnology, and relates to an application of NF-YC9 protein in regulating plant tolerance to ABA. Background technique [0002] Abscisic acid (ABA) is one of the important hormones in plants. It plays an important role in all stages of plant growth and development, including seed dormancy, germination, seedling growth, stomatal movement, and transition from vegetative growth to reproductive growth, etc. process. At the same time, ABA plays an important role in the response of plants to various external stresses, such as abiotic stresses such as drought, cold, high temperature, salt and mechanical damage, and biotic stresses such as pests and diseases. Therefore, the elucidation of the molecular mechanism of the plant ABA signal transduction pathway is of great significance for regulating plant growth, improving crop quality, improving genetic characteristics, and promoting the development of modern agricultural...
