Through carrying out smashing detection on a pollen sample, the method comprises the following steps of (A) preparing wall-broken pollen: acquiring non-broken pollen, putting into a wall breaking processor for smashing, and preparing to obtain quantitative wall-broken pollen; (B) preparing pollen mixed liquor: re-acquiring non-broke pollen in the same batch of the pollen in the step (A), wherein the mass of the non-broken pollen is the same as the mass of the prepared wall-broken pollen; respectively adding solvents with the viscosity being P into the non-broken pollen and the wall-broken pollen, compounding into non-broken pollen mixed liquor and wall-broken pollen mixed liquor with the same concentration,and standing after shaking up; (C) determining cells of the pollen mixed liquor: respectively sucking up the non-broken pollen mixed liquor and the wall-broken pollen mixed liquor with the same volume, and observing and calculating the number of whole cell contained in unit mass.
