Abstract

The invention discloses a lemon eucalyptus seed seedling method, which comprises the following steps of picking lemon eucalyptus fruits in autumn, separating seeds from seed coats, and performing coldstorage; soaking the seeds with a formalin solution firstly, then cleanly washing the seeds with clear water, and soaking the seeds with a chitosan-oligosaccharide solution to obtain treated seeds; laying the treated seeds on the bottom of a nursery bed by using gauze, uniformly covering the nursery bed with a nursery bed substrate, uniformly broadcasting the soaked lemon eucalyptus seeds on thenursery bed, and then covering the seeds with 1-2cm nursery bed substrate; after germination of the lemon eucalyptus seeds, controlling the temperature, the humidity and the illumination, obtaining lemon eucalyptus seedlings after germination of main leaves, transplanting the lemon eucalyptus seedlings into a nonwoven fabric mesh bag which is charged with a seedling substrate, putting the nonwovenfabric mesh bag in a plastic seedling tray, profoundly sprinkling water, and putting the plastic float tray in a shading shed; and sprinkling a salicylic acid solution and the chitosan-oligosaccharide solution once on the leaf surfaces of the lemon eucalyptus seedlings every 5-7 days. The method provided by the invention has the advantages of high germination rate, good reliability, no disease, greenness, simpleness in operation, low cost and high efficiency and the like and can realize mass propagation of the lemon eucalyptus seedlings.

Example Embodiment

[0025] Example 1
[0026] On November 6, 2015, the lemon eucalyptus fruit with full particles was picked, placed in a ventilated place in the room until the seeds fell off naturally, and then sieved with a sieve with an aperture of 4mm. The seeds were separated from the seed coat by a small windmill, and the seeds were placed at 0 Chilled in a cold storage at ℃ for later use; on September 25, 2016, the frozen seeds were soaked in a 0.15wt% formalin solution for 30 minutes, then rinsed with water, and then soaked in a chitosan solution with a concentration of 100mg/L for 24h , The seeds must be treated;
[0027] Mix the yellow heart soil and river sand evenly according to the mass ratio of 2:1 to obtain the seedbed substrate; select a smaller seedbed container, first spread the seedbed with gauze to the bottom of the seedbed, and evenly cover the seedbed with a thickness of 2-2.5 cm, on September 26, 2016, evenly spread the soaked lemon eucalyptus seeds to the seedbed, and then cover the seedbed substrate with a thickness of 1-1.5cm, and set up a 2m high shade shed on the seedbed, and the shade shed was covered The seedbed is equipped with a spray facility; the lemon eucalyptus seeds began to germinate on September 29, 2016, and the germination is basically complete after 2 days. After the germination starts, the temperature is controlled at 20-25℃ and the humidity is 85-95%. The true leaves began to germinate on the 4th, and lemon eucalyptus seedlings were obtained. According to statistics, the seedling rate of the lemon eucalyptus nursery was 90%.
[0028] On October 4th, the lemon eucalyptus seedlings were transplanted into a non-woven mesh bag with a diameter of 4 cm containing a seedling substrate. The non-woven mesh bag was placed in a plastic seedling tray, and then water was poured in, and the plastic seedling tray was placed In the shade shed, spray frequently to keep the seedling growing substrate moist; the seedling growing substrate selects fermented bran and bark according to the mass ratio of 7:3 and mixes evenly, and adds 2g/m 3 Slow-release fertilizer; spray salicylic acid solution and chito-oligosaccharide solution on the leaves of lemon eucalyptus seedlings every 7 days. The concentration of the chito-oligosaccharide solution is 100 mg/L, and the concentration of the salicylic acid solution is 10 mg/L. On December 26, the survival rate of lemon eucalyptus seedlings was calculated to be 90%. The average seedling height was 13cm and the main root length was 19.7cm.
[0029] 1. Investigate the effect of soaking with different concentration of chitosan on the germination of lemon eucalyptus seeds. The other test conditions are the same as this example. The effects of different treatments on the germination of seeds are shown in Table 1:
[0030] Table 1: The effect of different concentrations of chitooligosaccharides on the germination of lemon eucalyptus seeds
[0031]
[0032] Note: Different letters indicate that different treatments have significant effects on seed germination parameters (P <0.05)
[0033] From the above investigation results, it is known that soaking lemon eucalyptus seeds with chito-oligosaccharides can shorten the germination time and increase the germination rate. Different concentrations of chito-oligosaccharides also have a greater impact on the germination of lemon eucalyptus seeds, with lower or higher concentrations. High is not conducive to the germination of seeds. The best germination conditions of the chito-oligosaccharide seeds of Eucalyptus lemon were selected with a concentration of 100-200 mg/L.
[0034] 2. Investigate the growth of lemon eucalyptus seedlings under different seedling substrates. Other test conditions are the same as this example. The results of the investigation are shown in Table 2:
[0035] Table 2: Growth of lemon eucalyptus seedlings under different seedling substrates
[0036]
[0037]
[0038] Note: Different letters indicate that different treatments have significant effects on seedling growth (P <0.05)
[0039] From the above investigation results, it is known that different seedling substrates have a greater impact on the growth of lemon eucalyptus seedlings. The seedling substrate of this scheme is adopted (the seedling substrate is fermented bran and bark mixed evenly according to the mass ratio of 7:3, and Compared with yellow heart soil, lemon eucalyptus seedlings grow better when added with a slow-release fertilizer of 2g/m3.
[0040] 3. Investigate the growth of different concentrations of chito-oligosaccharide solutions sprayed on lemon eucalyptus leaves. Other test conditions are the same as this example. The growth of 90-day-old lemon eucalyptus seedlings is shown in Table 3:
[0041] Table 3: Growth status of different concentrations of chito-oligosaccharide solutions sprayed on lemon eucalyptus leaves
[0042]
[0043] Note: Different letters indicate that different treatments have significant effects on seedling growth (P <0.05).
[0044] From the above investigation results, it is known that spraying lemon eucalyptus leaves with different concentrations of chitosan solutions can promote the growth of lemon eucalyptus seedlings, but different concentrations of chitooligosaccharides also have a greater impact on the growth of lemon eucalyptus seedlings. Low or high is not conducive to the growth of seedlings. After experiments, the chito-oligosaccharide solution with a concentration of 100-200 mg/L was used to spray lemon eucalyptus seedlings, and the growth of the seedlings was the best.
[0045] 4. Investigate the growth situation of spraying salicylic acid solutions of different concentrations on the leaf surface of lemon eucalyptus. The other test conditions are the same as this example. The growth situation of lemon eucalyptus seedlings grown for 90 days is shown in Table 3:
[0046] Table 4: Growth situation of different concentrations of salicylic acid solution sprayed on lemon eucalyptus leaves
[0047]
[0048] Note: Different letters indicate that different treatments have significant effects on seedling growth (P <0.05).
[0049] From the above investigation results, it is known that spraying lemon eucalyptus leaves with different concentrations of shell salicylic acid solutions can increase the survival rate of lemon eucalyptus seedlings, but salicylic acid has little effect on the growth of lemon eucalyptus seedlings. Not conducive to the growth of seedlings. After testing, spraying lemon eucalyptus leaf seedlings with salicylic acid at a concentration of 100 mg/L showed the best growth and survival rate of the seedlings.
