Abstract
The invention discloses a medical plant black false hellebore immature embryoin vitro culture method which comprises the following operation steps of (1) collecting immature capsules of black false hellebore, and poking immature seeds out of capsules for sterilization treatment; (2) poking immature embryos in the seeds with a scalpel under an anatomical lens, and then putting the immature embryosin a soak solution for soaking treatment for 30-40 min; (3) adding the immature embryos treated in step (2) in an immature embryo culture medium for dark culture at 24-27 DEG C; and (4) when the immature embryos grow into young seedlings with the height being 4-5 cm, transferring the young seedlings into a rooting culture medium, and transplanting the seedlings into turfy soil after roots of theseedlings are well developed, wherein the rooting culture medium is prepared from an MS culture medium, 60-70 mg / L ferric salt and 0.8-1.2 mg / L indolebutyric acid, 2-5 mg / L sodium hyaluronate. The medical plant black false hellebore immature embryoin vitro culture method provided by the invention is simple to operate and can culture the immature embryos into the seedlings with high survival rate,wherein the average survival rate of the immature embryos can reach about 93 percent, and the average rooting rate of young seedlings can reach about 95 percent.
