4 results about "Sonic hedgehog" patented technology

Sonic hedgehog is a secreted protein that in humans is encoded by the SHH gene. It is the best studied ligand of the hedgehog signaling pathway, others being desert hedgehog (DHH) and Indian hedgehog (IHH). It plays a key role in the development of animals, from insects to mammals. In vertebrates, it is involved in organogenesis, including the growth of digits on limbs and the organization of the brain. Sonic hedgehog is an archetypal example of a morphogen as defined by Lewis Wolpert's French flag model—a molecule that diffuses to form a concentration gradient and has different effects on the cells of the developing embryo depending on its concentration. Sonic hedgehog is also active in adults; for example, it controls the proliferation of adult stem cells and has been implicated in the development of some cancers.

Spinal cord organoid and method for producing same

PCT designated stageWO2026049008A1Nervous disorderMicrobiological testing/measurementPluripotential stem cellRetinoic acid receptor
Provided are: a method for producing a spinal cord organoid from a pluripotent stem cell in vitro, the method comprising (1) a step for culturing a human pluripotent stem cell in a culture medium that contains a ROCK inhibitor and a Wnt signal activator but does not substantially contain a TGFβ inhibitor or a BMP signal inhibitor, (2) a step for further culturing the cell obtained in step (1) in a culture medium that contains a retinoic acid receptor agonist and a hedgehog signal activator, and (3) a step for further culturing the cell obtained in step (2) in a culture medium that does not substantially contain a TGFβ inhibitor, a BMP signal inhibitor, a retinoic acid receptor agonist, or a sonic hedgehog stimulator to obtain a spinal cord organoid; and a spinal cord organoid obtained by the method. The spinal cord organoid according to the present application comprises at least a portion of the dorsal side of the spinal cord and a ventral region of the spinal cord.

Method for producing dopaminergic neuron progenitor cell

PendingUS20260176580A1Culture processNervous system cellsCell-Extracellular MatrixDifferentiation-inducing factor
A cell population comprising Corin- and / or Lrtm1-positive cells was produced by the following steps (1) and (2), from which Corin positive and / or Lrtm1 positive cells are collected using a substance that binds to Corin and / or a substance that binds to Lrtm1, and dopaminergic neuron progenitor cells are produced by performing suspension culture of the Corin positive and / or Lrtm1 positive cells in a culture solution containing one or more nutritional factors:(1) a step of performing adhesion culture of pluripotent stem cells in a medium for maintaining undifferentiated state containing a Sonic hedgehog (SHH) signal stimulant, and an undifferentiated state-maintaining factor in the absence of feeder cells but in the presence of an extracellular matrix, and(2) a step of culturing the cell population obtained in the step (1) in a culture solution containing one or more differentiation-inducing factors.

Method for producing cell mass including pituitary tissue, and cell mass thereof

The present invention aims to provide a method for efficiently producing a cell mass containing pituitary tissue from pluripotent stem cells. A method for producing a cell mass containing pituitary tissue, including the following steps (1) and (2):(1) a first step of suspension-culturing pluripotent stem cells to form a cell aggregate in the presence of a Wnt signal transduction pathway inhibiting substance,(2) a second step of suspension-culturing the aggregate obtained in the first step in the presence of a BMP signal transduction pathway activating substance and a Sonic hedgehog signal transduction pathway activating substance, thereby obtaining a cell mass comprising pituitary tissue.
