The floor plate is a structure integral to the developing nervous system of vertebrate organisms. Located on the ventral midline of the embryonic neural tube, the floor plate is a specialized glial structure that spans the anteroposterior axis from the midbrain to the tail regions. It has been shown that the floor plate is conserved among vertebrates, such as zebrafish and mice, with homologous structures in invertebrates such as the fruit fly Drosophila and the nematode C. elegans. Functionally, the structure serves as an organizer to ventralize tissues in the embryo as well as to guide neuronal positioning and differentiation along the dorsoventral axis of the neural tube.

The present application provides a method for producing, from pluripotent stem cells, neural progenitor cells of the midbrain floor plate region or dopaminergic neural progenitor cells, the method comprising (1) inducing differentiation of pluripotent stem cells into neural progenitor cells of the midbrain floor plate region, and optionally (2) inducing differentiation of the neural progenitor cells of the midbrain floor plate region, which are obtained by the differentiation induced in (1), into dopaminergic neural progenitor cells, wherein all or part of the differentiation induced in (1) includes culturing the cells in a medium containing an agonist for LPA receptor.

Methods of differentiating neural cells and related compositions and methods of use
The present disclosure provides methods of lineage specific differentiation of pluripotent stem cells, including induced pluripotent stem cells, into floor plate midbrain progenitor cells, determined dopamine (DA) neuron progenitor cells, and / or DA neurons. Also provided are compositions and uses thereof, such as for treating neurodegenerative diseases and conditions, including Parkinson's disease.
