[0050]Isolation and Genotyping of Example 1 Lactobacillus plantarum subspecies PS128

[0051] Lactobacillus plantarum subsp. plantarum PS128 (hereinafter referred to as PS 128) was isolated from fu-tsai (a traditional fermented nitrogen mustard product in Taiwan) and cultured in a specific artificial medium. The 16S rRNA gene (SEQ ID NO. 3) from PS128 was analyzed by direct sequencing of about 1000 nucleotides of 16S rDNA amplified by PCR using primers represented by SEQ ID NO: 1 and SEQ ID NO: 2. Genomic DNA extraction, PCR-mediated amplification of 16S rDNA, purification of PCR products, and sequencing of purified PCR products were performed under the conditions indicated in Table 1.

[0052] The resulting sequence was imported into the comparison software provided by the National Center for Biotechnology Information (NCBI) (http: / / www.ncbi.nlm.nih.gov / ), manually compared and compared with the thick-walled Representative 16S rDNA sequences of organisms of the phylum Firmic...

[0064] Example 2 Using ERIC-PCR patterns to identify novel strains of Lactobacillus plantarum subspecies PS128

[0065] ERIC-PCR was performed to further distinguish bacterial subspecies with high sequence similarity. The ERIC-PCR profiles of Lactobacillus plantarum strains were performed under the conditions indicated in Table 3. DNA extracted from PS128 and two Lactobacillus plantarum strains were used as templates. Such as figure 1 As shown in , the resulting amplified products were subjected to electrophoresis and compared patterns using primers represented by SEQ ID NO: 4 and SEQ ID NO: 5.

[0073] Example 3 Analysis Type Index (API) Typing

[0074] The sugar utilization of PS128 used in the present invention was studied using API 50 CHL kit (bioMerieux, France), and the results are shown in Table 4. Fermentation tests showed that PS128 had biochemical properties similar to Lactobacillus plantarum subsp. plantarum.

Abstract

The invention surprisingly found that Lactobacillus plantarum subsp. plantarum PS 128 provides an advantageous effect in treatment or prevention of tic disorders and basal ganglia disorders. Accordingly, the invention provides a method of treating or preventing a movement disorder in a subject, comprising administering to a subject an effective amount of cells of a Lactobacillus plantarum subsp. plantarum PS128, which is deposited under DSMZ Accession No. DSM 28632.

Description

technical field [0001] The present invention relates to a method for preventing or treating movement disorders. Specifically, the present invention provides a method for preventing or treating tic disorders and basal ganglia disorders by using lactic acid bacteria. Background technique [0002] The basal ganglia contain various subcortical nuclei, including the striatum, pallidus, substantia nigra, and subthalamic nuclei, which are located at the base of the vertebrate forebrain. The basal ganglia nucleus is strongly interconnected with several other brain regions including the cortex, thalamus, and brainstem. Numerous studies have shown its involvement in various brain functions such as control of voluntary motor actions, procedural learning, routine behavior, eye movement, cognition and emotion. Multiple and diverse neurological conditions are associated with dysfunctional behavioral control of the basal ganglia, including hyperkinetic disorders such as unilateral twitch...
