[0132] The preparation of the miRNA chip can adopt conventional manufacturing methods of biochips known in the art. For example, if the solid phase carrier is a modified glass slide or silicon wafer, and the 5' end of the probe contains amino-modified poly dT strings, the oligonucleotide probe can be formulated into a solution, and then spotted with a spotting instrument. The miRNA chip of the present invention can be obtained by arranging in a predetermined sequence or array on a modified glass slide or a silicon chip, and then fixing it by standing overnight. If the nucleic acid does not contain amino modification, its preparation method can also refer to:

[0151] Strictly abide by the "Interim Measures for the Management of Human Genetic Resources" promulgated by the State Council in 1998 and the 2005 "Technical Regulations for the Collection, Arrangement and Preservation of Fresh Human Genetic Resources Disease Tissues" and take this as a benchmark. From January 2017 to February 2019, 78 patients with colorectal cancer, 70 patients with colorectal adenoma and 70 healthy patients were collected from the gastrointestinal surgery, endoscopy center, gastroenterology and physical examination center of the Third Affiliated Hospital of Guangzhou Medical University. The peripheral venous blood of the control group and the patients with colorectal cancer were all diagnosed by pathological examination, and did not include other tumor diseases or major and chronic diseases that may affect the diagnosis of the target of this study. All patients had complete clinical and pathological data. There were 43 males and ...

Abstract

The invention discloses a plasma marker combination for early diagnosis or prognosis prediction of colorectal cancer. Specifically, the present invention obtains miRNA which is obviously abnormally expressed in colorectal cancer plasma, namely miR-224-5p, miR-301a-3p, miR-145-5p, miR-193b-3p and miR-1183. The invention finds that the plasma miRNA combination can be used as a marker for early diagnosis or prognosis judgment of colorectal cancer by researching the expression difference of the plasma miRNA in patients with colorectal cancer and colorectal adenoma at various stages and healthy people.

Description

technical field [0001] The invention belongs to the field of biotechnology and medicine, in particular, the invention relates to a plasma marker combination for early diagnosis or prognosis prediction of colorectal cancer. Background technique [0002] Colorectal cancer (CRC) is one of the most common malignant tumors. From a global perspective, the incidence of CRC ranks second in women, third in men, and the overall CRC mortality rate ranks second. An estimated 1.8 million new cases of colorectal cancer and 881,000 deaths will occur in 2018, accounting for approximately 1% of all cancer cases and deaths. By 2030, the global incidence of CRC is expected to increase by 60%. In 2015, China's cancer statistics show that the incidence of CRC ranks fifth among males, fourth among females, and ranks fifth in both overall morbidity and mortality. The overall incidence of gastrointestinal tumors is increasing year by year. According to literature statistics, after standard treatm...
