Abstract

The invention provides a sesamum indicum unpollinated ovary in-vitro culture callus induction method. The method comprises the following steps: (1) material selection and disinfection: selecting a sesamum indicum plant which grows robustly and has no plant diseases and insect pests, collecting unpollinated flower buds of which anthers do not crack 1-2 days before flowering, and carrying out disinfection treatment; (2) explant pretreatment: stripping ovaries as explants, and soaking the explants in a vitamin C solution with a certain concentration for 2 hours; and (3) explant inoculation and induction culture: flatly placing and inoculating the explant on an induction culture medium, carrying out heat shock treatment for 24-48 hours, then carrying out induction culture at the culture temperature of 23-28 DEG C, carrying out dark culture for the first 30 days in the culture period, and then carrying out illumination culture. According to the method disclosed by the invention, the sesamumindicum unpollinated ovary callus is successfully induced, and the highest induction rate can reach 36.74%. The invention lays a material foundation for further establishing a complete sesamum indicum unpollinated ovary in-vitro culture system.
