Abstract

The method comprises the following steps of selecting a test strain and an alfalfa variety, preparing a test medium, extracting pantoea agglomerans lipopolysaccharide (LPSp), detecting the activity of the pantoea agglomerans lipopolysaccharide, and carrying outa tissue culture germination test. Germination test seeds are divided to be subjected to two kinds of treatment including disinfection and non-disinfection, a non-disinfection group is not subjected to any treatment, 1 ml, 2 ml and 3 ml of LPSp are added into glass tissue culture germination bottles in a treatment group correspondingly, and the concentrations of the lipopolysaccharide are respectively 0.267 EU/mL, 0.534 EU/mL and 0.801 EU/mL and are marked as A, B and C for treatment; and sterile water treatment is used as a control group. According to the method, 0.267 EU/mL of LPSp promotesnodulation of alfalfa roots of the non-disinfected group, 0.267 EU/mL of LPSp accelerates nodulation of the alfalfa roots of the non-disinfected group, and nodulation is about 18 days earlier than that of the control group; 0.267 EU/mL of LPSp does not affect seed germination including germination potential and germination index; and along with the increase of the concentration of the LPSp, the germination and growth of the alfalfa seeds are inhibited, and the germination and growth of the seeds in the disinfection group are completely inhibited by 0.801 EU/mL of LPSp.
