[0126] Detection and identification of HBV surface antigen mutants in Singaporean adults and vaccinated minors with high anti-HBs levels but surface antigen negative

[0127] Using the primer oligonucleotides provided by the present invention, whether there is HBV DNA in HBsAg-negative Singaporean adults and vaccinated minors was studied by polymerase chain reaction (PCR) amplification. A total of 63 adults and 15 vaccinated minors (less than 15 years old, vaccinated with recombinant vaccines) from different ethnic minority groups were selected and all were negative for HBsAg by 4 independent immune-based commercial diagnostic kits (Table 2). All of them were positive for total antibody to HBV core antigen (anti-HBc) (Corzyme, Abbott Laboratories, USA). All were also positive for anti-HBs (Ausab, Abbott Laboratories, USA) (Table 2). DNA was extracted from 100 [mu]l serum by proteinase K treatment, followed by chloroform / phenol extraction and ethanol precipitation. Oligonucl...

Abstract

The present invention relates generally to a nucleic acid-based assay to detect the presence of a viral pathogen and, in particular, hepatitis B virus. More particularly, the present invention provides a single-step amplification assay to detect hepatitis B viral nucleic acid sequences. The test of the present invention is readily adaptable for automation and permits the rapid through-put of samples to be tested. The present invention further provides agents useful for performing a nucleic acid-based detection assay for hepatitis B virus and a kit comprising said agents.

Description

technical field [0001] The present invention generally relates to nucleic acid-based assays that can be used to detect the presence of viral pathogens, particularly hepatitis B virus. More particularly, the present invention provides a one-step amplification assay for detecting nucleic acid sequences of hepatitis B virus. The assay of the present invention can be easily adapted for automation and allows rapid flow through of the samples to be tested. The present invention also provides a reagent and a kit containing the reagent, and the reagent can be used for nucleic acid-based hepatitis B virus detection test. Background technique [0002] A detailed bibliography of the publications referred to numerically in the specification is centrally located at the end of the specification. [0003] Hepatitis B virus (hereinafter "HBV") can cause a debilitating disease state and can lead to acute liver failure, cirrhotic hemorrhage and primary liver cancer. HBV infects millions of...
