[0099] 5.6 Preparation of animal feed

[0100] Polypeptides of the invention prepared as described above in Sections 5.1-5.5, which have phytase activity at neutral pH, are useful in providing animal feeds in which phosphorus is efficiently utilized by animals raised on such feeds. Accordingly, another aspect of the invention is to provide animal feed comprising a polypeptide of the invention which releases phosphorus from phytate for use by the animal. For example, such animal feed preparation method can be: the feed is mixed with the phytase powder of the present invention and made into pellets, wherein the enzyme powder has an enzyme activity of 200,000-400,000 EU at a weight ratio of about 1 kg of powder per ton of feed / kg. In order to mix evenly, the enzyme powder can be mixed with a small amount of feed such as 10 kg first, and then mixed with the rest of the feed. The amount of enzyme used in the feed is at least 200 enzyme units (EU) / kg feed, preferably at least 250...

Abstract

The present invention clones and analyzes two phytase genes from microorganisms recognized as safe: Bacillus licheniformis and Bacillus subtilis 168. A method for overexpression and purification of phytase was established. These enzymes have a molecular weight of approximately 48 kDa (kilodaltons) and possess extracellular phytase hydrolytic activity. These recombinant enzymes can be used to enhance phytases in various commercial applications, including the production of animal feed and transgenic plants with increased rates of mature growth, flowering and fruiting.

Description

[0001] This application benefits from and claims priority to US Provisional Application Serial No. 60 / 332,060, filed November 21, 2001, which is hereby incorporated by reference in its entirety. [0002] 1 Introduction [0003] The present invention relates to phytase genes from two generally recognized as safe (GRAS) microorganisms, Bacillus licheniformis and Bacillus subtilis 168, and their respective encoded protein products and fragments, derivatives, analogs and variants thereof. The present invention also provides methods of producing and purifying phytases, derivatives, analogs, variants and antibodies. The invention also provides the use of these phytases in animal feed. The invention also provides transgenic plants in which these two phytases are active at neutral pH ("neutral phytases") and other neutral phytases. These transgenic plants had increased growth, flowering and fruiting. 2. Background of the invention [0004] Phytate is the salt form of phytic acid (m...
