Embodiment 1

[0021] Preparation of colloidal gold pads

[0022] Prepare a colloidal gold solution with a diameter of 15nm by the chloroauric acid-trisodium citrate reduction method. After the preparation is completed, take 100ml of the colloidal gold solution in a beaker and use 0.1M K 2 CO 3 Adjust the pH to 8.0, add 0.65 mg mouse anti-human IgM antibody (Shanghai Lingchao Biotechnology Co., Ltd.) that can specifically bind to the IgM antibody of HSV-I and HSV-II per 100 ml of colloidal gold solution, stir at room temperature for 1 hour, add 5% bovine serum albumin (BSA) by weight to block for 1 hour, centrifuge at 12,000 rpm and 4°C for 30 minutes, discard the supernatant, redissolve to 100ml with colloidal gold solution, and spread 20cm by 1ml solution 2 Spread the solution evenly on the glass fiber membrane and dry at 37°C for 30 minutes to make a colloidal gold pad.

Abstract

The invention discloses a test strip for quickly detecting antibodies in saliva for determining herpes simplex viruses, which comprises a bottom plate. A sample pad, a colloidal gold pad, a nitrocellulose membrane and a suction pad are sequentially pasted on the bottom plate; a mouse anti-human IgM antibody which is marked with colloidal gold and can be specifically combined with IgM antibodies ofHSV-I and HSV-II is attached to the colloidal gold pad; and a detection line and a quality control line are coated on the nitrocellulose membrane, wherein the detection line comprises human recombinant HSV-I and HSV-II antigens, and the quality control line can be specifically combined with the mouse anti-human IgM antibody and comprises sheep anti-mouse anti antibodies. The test strip of the invention is used for detecting the IgM antibodies of HSV-I and HSV-II in the saliva by adopting the immunochromatography technique, thereby determining whether people are infected with herpes simplex viruses or not. The test strip has the characteristics of quick reaction speed, high sensitivity, strong specificity, simple operation and lower detection cost and is suitable for self detection.

Description

technical field [0001] The invention relates to the technical field of biological diagnosis, in particular to a test strip for rapid detection of herpes simplex virus antibody saliva. Background technique [0002] Herpes simplex is caused by Herpes Simplex Virus (HSV), a DNA virus. Human herpes simplex virus is divided into two types, namely herpes simplex virus type I (HSV-I) and herpes simplex virus type II (HSV-II). Type I mainly causes infections of the skin, mucous membranes (oral mucosa) and organs (brain) other than the genitals. Type II mainly causes skin and mucous membrane infection in the genital area. These two types can be identified by fluorescent immunoassay and cell culture method. [0003] Humans are the only natural host of HSV. The virus enters the body through the respiratory tract, oral cavity, genital mucous membrane and damaged skin, and resides in the normal mucous membrane, blood, saliva and sensory ganglion cells of the human body. Most of the ...
