The manufacturing method comprises the following steps of: preparing a suspension sizing agent by using water, the cuttlefish bone meal and the clay, and heating the suspension sizing agent at the temperature of 15 DEG C until the suspension sizing agent is boiling; immersing the substrate material into the sizing agent, or coating the sizing agent on the substrate material; taking the substrate material impregnated or coated with the sizing agent out, and performing flat pressing or rolling, or after taking the substrate material impregnated or coated with the sizing agent out, covering one side or both sides of the substrate material coating with the sizing agent by using medical cotton yarn or non-woven fabrics, and performing the flat pressing or the rolling again; and performing drying and sterilization to obtain the clay-enhanced cuttlefish bone meal hemostatic gauze.
