Embodiment Construction

[0020] Concrete processing steps of the present invention are as follows:

[0021] 1. Compound enzyme extraction process steps:

[0022] The main steps are: raw material processing → bromelain hydrolysis to remove non-fibrous proteinwater washing → 537 acid protease hydrolysis residue collagen → normal pressure filtrationfreeze dryingdry storage.

[0023] 1.1 Raw material pretreatment

[0024] Thaw and wash the fish leftovers (including fish skin, fish bones, and fish scales), crush them into small pieces of tissue with a pulverizer, degrease (soak in 2.5% NaCl solution at 5°C for 10-20h), centrifuge (6000r / min 5 ℃ low temperature centrifugation for 30min), remove the waste liquid, and obtain the leftovers after pretreatment, as the raw material for collagen protein extraction.

[0025] 1.2 Enzymolysis

[0026] The factors affecting the effect of enzymatic hydrolysis mainly include: the type of enzyme, the amount of enzyme added, the temperature of enzymatic hydrol...

Abstract

The invention discloses a method for extracting collagen from fish processing waste including skin, bones and scale. The method mainly comprises the following steps of: processing raw materials; hydrolyzing by using bromelin to remove non-fibrous proteins; washing with water; hydrolyzing residual collagen by using 537 acid prolease; filtering under normal pressure; performing freeze drying; and drying and storing. Keys points of the method lie in enzymolysis and purification of the collagen. A higher-purity product can be prepared from fish leftover collagen extracted with the method; and the product is white, does not have any peculiar smell, and has the average extraction rate of 38.9 percent and the purity of 99.67 percent.

Description

Technical field: [0001] The invention relates to a method for extracting collagen, in particular to a method for extracting collagen from skin, bone and scale of fish processing waste. Background technique: [0002] my country has vast waters and abundant aquatic resources. With the development of the aquatic product processing industry, a large amount of leftovers are produced while processing fish products, which account for 50% to 70% of the total mass of processed fish. However, restricted by technology and traditional concepts, for a long period of time, aquatic waste was not fully utilized, but was thrown away as garbage. It is estimated that the total amount of fish waste in my country reaches more than 2 million tons every year. At present, fish leftovers in my country are mainly used for: (1) processing into feed fish meal; (2) extracting fish collagen; (3) processing into fish maw; (4) extracting fish oil from fish viscera, refining EPA and DHA products.
