Problems solved by technology

However, red ginger is mainly propagated through asexual ramets, which has weak meristematic ability, low reproductive coefficient in the natural state, and is not resistant to transplantation, which is far from meeting the needs of large-scale production; the fresh-cut flower arrangement period of red ginger is only 3 to 5 days , the plant is as high as 2 meters, which also limits its commercial value as an ornamental flower
So far, there are no reports on the establishment of ECS and high-frequency somatic embryogenesis of plants regenerated by ECS.

Embodiment 1

[0063] The method for establishing the high-efficiency somatic embryogenesis and regeneration plant of red ginger flower comprises the following steps:

[0064] (1) Selection of ginger flower species: The selected species is red ginger flower (Hedychium coccineum Buch.-Ham) with dense flowering, bright flowers, long flowering period, which can be directly used for garden layout and flower breeding materials. From Panyu Experimental Base of Zhongkai Agricultural Engineering College;

[0065] (2) Induction of callus and optimized proliferation of embryogenic callus: From mid-June to mid-July, the immature inflorescence florets of Red Ginger were washed with tap water for 30 minutes, and then washed with 0.1wt% HgCl 2 Disinfect for 10 minutes, rinse 5 times with sterile water; peel off the outer bracts on the ultra-clean workbench, cut the anthers and filaments into 0.5cm sections, inoculate them on the callus induction medium, and store them in the dark at 28±1°C Cultivate under ...

Abstract

The invention discloses a method for efficiently regenerating plant through Hedychium coccineum Buch.-Ham somatic embryogenesis. The method comprises the following steps of: inducing calluses by using immature filaments and anthers of Hedychium coccineum Buch.-Ham as explants, conducting optimization, screening and subculture to obtain high-quality embryonic calluses, conducting suspension culture to the embryonic calluses and establishing an embryonic cell suspension culture system of Hedychium coccineum Buch.-Ham. A great number of somatic embryos can be obtained by using embryonic cell suspension cultures for inducing somatic embryos, an optimum effect can be obtained by using SH inorganic salt and 150mug.L<-1> TDZ, and the somatic embryo generation rate can reach 4429 / ml SCV (settled cell volume); the obtained somatic embryos can germinate seedlings with buds and roots on somatic embryo germination culture medium; and when SH culture medium is used as basic culture medium and the concentration of 6-BA is 0.5-1.0mg.L<-1>, the germination rate of the somatic embryos can reach 100 percent. Due to the method, a foundation is laid for the in-vitro rapid propagation and the biotechnological breeding of Hedychium coccineum Buch.-Ham, the large-scale popularization and application are facilitated and the application prospect is better.

Description

technical field [0001] The invention belongs to the field of biotechnology breeding of Zingiberaceae (Zingiberaceae) genus Hedychium, and particularly relates to a method for efficiently regenerating plants through somatic embryogenesis pathway of red ginger flower (Hedychium coccineum Buch.-Ham). Through this method, the embryogenic cell suspensions (ECS) of Red Ginger Flower can be established, and the high-efficiency somatic embryogenesis frequency can be obtained from the embryogenic cell suspension culture line, and an efficient plant regeneration system can be successfully established. Background technique [0002] Red ginger flower (Hedychium coccineum Buch.-Ham) is a perennial herb of Zingiberaceae Zingiberaceae (Hedychium), distributed in my country's Tibet, Yunnan and Guangxi, and along the Himalayas, Sri Lanka, Myanmar and Vietnam. Red ginger flower is a treasure in the genus Ginger, with high ornamental value. The bright red spikes are suitable for high-end fresh...
