Embodiment 1

[0017] Embodiment 1: Preparation of Sudan Grass S722 protoplasts

[0018] 1. Preparation of sudan grass seedlings: select plump sudan grass S722 seeds, plant them in a nutrient bowl, and cultivate them at 25°C for 8 days.

[0019] 2. Treatment of young leaves and stems: cut 1 g of young leaves and stems of sudan grass S722, and cut the leaves into about 1 mm length with a blade.

[0020] 3. Vacuum enzymatic hydrolysis: put the cut stems and leaves into the triangular flask with 10mL of enzymatic hydrolysis solution, and put it in a vacuum environment (-0.1MPa) for 1 hour. After taking it out, cover the mouth of the triangular flask with tin foil ℃ shaker, rotating speed 40rpm, enzymatic hydrolysis for 3 hours. (enzymolysis solution formula: 0.645M mannitol, 10mM potassium chloride, 1mM calcium chloride, 8mM 2-morpholineethanesulfonic acid, pH5.5, 1.5% cellulase and 0.3% isolated enzyme R-10).

[0021] 4. Dissociation of protoplasts: Pour off the enzymatic solution, add 10ml ...

Abstract

The invention discloses a technique for preparing a Sudan grass protoplast by using a vacuum enzymolysis method, belonging to the field of biotechnology. The technique comprises the steps of: chopping spires and stems of the Sudan grass in a certain size after the Sudan grass seeds sprout; and performing enzymolysis on cell walls of both the spires and the stems by utilizing the vacuum enzymolysis method so as to separate out a plenty of Sudan grass protoplasts. By utilizing the method, a plenty of Sudan grass protoplasts with complete cells can be prepared conveniently and rapidly.

Description

technical field [0001] A vacuum enzymolysis method for preparing protoplasts of Sudan grass belongs to the field of biotechnology, more specifically, it is a technology for preparing a large amount of protoplasts by using cell wall lyase to crack the cell walls of young leaves and stems of Sudan grass in a vacuum environment. Background technique [0002] Protoplast refers to a cell from which the cell wall has been removed or a naked cell surrounded by a plasma membrane. Mass preparation of protoplasts is an important basis for cell tissue culture and plant gene function research. The preparation method of Sudan grassland protoplasts has not been reported yet, and the establishment of the preparation method of Sudan grassland protoplasts is conducive to the further development of related research on Sudan grass. [0003] Contents of the invention [0004] The purpose of the present invention is to adopt a method of vacuum enzymolysis to prepare a large amount of protopl...
