specific Embodiment approach 1

[0016] Specific embodiment one: the method for improving Solanum nigrum seed germination in the present embodiment carries out according to the following steps:

[0017] 1. Collection and selection of seeds: From August 1st to September 10th every year, collect Solanum nigrum seeds, wash them with running water for 5 minutes, and let them dry naturally at room temperature and relative humidity of 30% to 50%. Dry for 3 to 5 months, then disinfect;

[0018] 2. Place the Solanum nigrum seeds treated in step 1 in the germination bed of a petri dish with a diameter of 60 to 90 mm, then seal the bottom of the petri dish and the junction of the cover with a breathable and moisturizing sealing film. Under the condition of 50-100%, take 24 hours as a cycle, culture for 10-15 days, each cycle at high temperature and each cycle at high temperature 20-28 ° C under light for 12 hours, and then at low temperature 10-16 ° C Cultivate under dark conditions for 12 hours, and the temperature d...

specific Embodiment approach 2

[0019] Specific embodiment two: the difference between this embodiment and specific embodiment one is that the disinfection method described in step one is as follows:

[0020] Solanum nigrum seeds dried naturally are put into 0.1% mercuric chloride solution for immersion and disinfection for 5 to 10 minutes, and then washed with distilled water for 3 to 5 times. Others are the same as in the first embodiment.

specific Embodiment approach 3

[0021] Embodiment 3: The difference between this embodiment and Embodiment 1 or 2 is that the germination bed of the culture dish described in step 2 is peat soil, fine river sand or qualitative filter paper. Others are the same as those in the first or second embodiment.

Abstract

The invention provides a method for improving seed germination of solanum nigrum, which relates to a method for improving seed germination. The method provided by the invention overcomes the technical problem of a low germination rate of the seed of solanum nigrum. The method comprises the following steps: 1, collecting and selecting seeds; and 2, putting the solanum nigrum seeds treated in step 1 into a germinating bed of a culture dish with a diameter of 60 to 90 mm, sealing an intersection part of the bottom and the cover of the culture dish by using a ventilating moisture-retaining sealing film and carrying out culture for 10 to 15 d under the condition of humidity of 50 to 100% with 24 h as a cycle period, wherein in each cycle period, the solanum nigrum seeds are cultured for 12 h at a high temperature of 20 to 28 DEG C and then cultured for another 12 h at a low temperature of 10 to 16 DEG C, and temperature difference between the high temperature and the low temperature in each cycle period is no less than 10 DEG C. The method provided by the invention is simple and easily practicable and enables the germination rate of the solanum nigrum seed to be increased to 57 to 63%, which is substantially higher than a germination rate obtained at a constant temperature of 28 DEG C.

Description

technical field [0001] The present invention relates to a method for improving seed germination. Background technique [0002] Solanum nigrum (Solanum nigrum) is a dicotyledonous plant, Solanaceae Solanum genus annual herb, widely distributed in my country, is a kind of wild plant resources, planting pearls and fruits can be used as medicine, the drug value is very high; the pollution of heavy metal cadmium Soil has a strong ability to repair; in addition, Solanum nigrum has certain characteristics of salt-alkali tolerance, and it is also a component species of vegetation in saline-alkali land. At present, most of the researches on Solanum nigrum focus on the extraction and utilization of its medicinal ingredients, and its role in the process of soil remediation. There are relatively few studies on its seed dormancy and germination characteristics, and promoting the germination of Solanum nigrum seeds is the key to improving its remediation potential. One of the technologies...
