Embodiment 1

[0033] The cloning of embodiment 1HPSE gene

[0034] (1) Primer design

[0035] The primer design work refers to the basis of the applicant’s previous work (patent document publication number: CN101892225A, publication date: 2010.11.24, invention name: HPSE gene as a molecular marker related to pig immune traits and its application, patent number: ZL 201010223062.0), employing HPSE gene cDNA (GenBank accession number: NM_006665.3) was used as an information probe. Using the BLAST tool in NCBI to screen homologous sequences in the GenBank pig EST database, a series of ESTs with more than 80% homology (fragment length greater than 100bp), use ENTREZ (http: / / www.ncbi.nlm.nih.gov / Web / Search / index.html) to query the corresponding sequences of these ESTs in NCBI, and then use the ASSEMBLY program in GeneTool to construct Pig EST-contigs. A pair of primers were designed according to the EST splicing sequence, and its nucleotide sequence is as follows:

[0036] HPSE: forward primer...

Embodiment 2

[0042] Embodiment 2. Fluorescence quantitative detection of HPSE gene mRNA expression in placental tissue

[0043] (1) Collection of porcine chorionic tissue samples

[0044] Purebred Meishan pigs and Large White young sows (from the boutique pig farm of Huazhong Agricultural University) were selected as the research objects, and a purebred Meishan boar (from the boutique pig farm of Huazhong Agricultural University) was mated with Meishan sows through artificial insemination. A purebred Large White boar was mated with a Large White sow (from the boutique pig farm of Huazhong Agricultural University) for two consecutive days. On the 26th day, 50th day, 95th day and 114th day of gestation (two pregnant sows of each breed (Meishan pig and Large White gilt) in each period) were slaughtered to collect chorionic tissue samples of pig placenta .

[0045] (2) Extraction of tissue total RNA

[0046] Total RNA was extracted from endometrial tissue samples of Meishan and Large White ...

Embodiment 3

[0065] Example 3 Detection of in situ expression and localization of HPSE gene mRNA in pig placenta tissue

[0066] (1) Sample collection

[0067] Pig placental tissue samples from Large White pigs on the 26th and 51st days of pregnancy and Meishan pigs on the 95th day of pregnancy were collected, cut into 1cm×2cm tissue pieces, and soaked in 4% paraformaldehyde solution for 24h-48h.

[0068] (2) Paraffin section production

[0069] 1) Washing with water: place the pig placenta tissue obtained in the previous step in a dehydration box, and wash with running water for about 12 hours.

[0070] 2) dehydration: the pig placental tissue after the last step process is placed in the dehydration box and uses gradient concentration (50% (2h), 70% successively, (overnight) 80% (2h), 95% (2h), 100%I (2h), 100% II (2h)) alcohol dehydration to 100% absolute ethanol;

[0071] 3) Transparency: The pig placenta tissue treated in the previous step was transparentized with 1:1 (volume ratio)...

Abstract

The invention belongs to the technical field of pig molecular marker preparation and application, and specifically relates to a novel purpose of a molecular marker related to reproductive traits and especially a pig birth weight trait. The molecular marker is cloned from HPSE gene. The nucleotide sequence of the molecular marker is represented by SEQ ID NO: 1. An A\G allele mutation is at 80bp of the sequence, such that Alu I-RFLP polymorphism is caused. The invention also discloses steps of the specific novel application of the molecular marker.

Description

technical field [0001] The invention belongs to the technical field of preparation and application of molecular markers for pigs, and in particular relates to the application of molecular markers for pig reproduction-related traits, especially pig birthweight traits. The molecular marker is related to the trait of pig birth weight, and is cloned from the HPSE gene. The invention mainly relates to a new application of the molecular marker. Background technique [0002] Pig is an important economic animal, as one of the main sources of animal protein for the majority of people in our country. In recent years, people's consumption of pork has been increasing day by day. For pig producers, how to improve production performance and reduce production costs has become the focus of their attention. In the past few decades, breeders have genetically improved many important economic traits of pigs through conventional breeding techniques, and achieved satisfactory results (such as le...
