The preparation method comprises the following steps of: extracting dry patrinia villosa root powder by using 75% ethanol under a room temperature and then carrying out ultrasonic extraction; standing still and filtering; distilling a filtering solution under reduced pressure to obtain rough extract; adding a proper amount of water into the rough extract to be diluted into a mixed suspension solution; extracting for three times by using petroleum ether, ethyl acetate and water saturated n-butyl alcohol respectively; mixing an n-butyl alcohol layer extracting solution; concentrating the n-butyl alcohol layer extracting solution under reduced pressure; then freez-drying to a constant weight to obtain an n-butyl alcohol extract for later use. The patrinia villosa root extract obtained by the preparation method provided by the invention has good capability of removing DPPH (Diphenylpicrylhydrazyl) free radicals and ABTS (2,2'-azino-bis(3-ethylbenzthiazoline-6-sulphonic acid)) free radicals.
