The method comprises the following processing steps: (1) taking crocodile oil fat, rejecting blood vessels and mucosal tissues, cutting the crocodile oil fat into strips, coarsely cutting the strips into particulate matters, putting the particulate matters into a beaker, adding a distilled water solution containing 2-8% of a citric acid, soaking and putting the mixture into a microwave oven to process for 5-10 minutes, carrying out secondary microwave treatment after cooling the mixture, and then continuously carrying out microwave low heat-assisted extraction on the mixture for 3-8 times; (2) adding a reactivating enzyme, adjusting the pH to be 4-6, putting the mixture into a bath pan to adjust temperature, stirring the mixture at a low speed, enhancing catalysis of an oil-water interface, and carrying out enzymolysis for 2-6 hours; (3) warming up the mixture to 85-95 DEG C after enzymolysis, inactivating the mixture for 10-20 minutes, and putting the mixture into a separating funnel to carry out oil-water separation, so as to remove moisture; (4) shaking and washing the crocodile oil for 3-5 times by using a chitosan distilled water liquid; and (5) refining the crocodile oil at 0-6 DEG C in winter, and centrifuging the crocodile oil at a high speed, so as to obtain the crocodile essential oil and crocodile essential oil fat.
