Embodiment Construction

[0015] Isolation and culture of bone marrow mesenchymal stem cells (MSCs):

[0016] The chicks were sacrificed by neck dislocation, the intact femur and tibia were aseptically separated, the attached muscle tissue was removed, and the bone marrow cavity was exposed and washed repeatedly, especially the metaphysis. The washed bone marrow cells were screened to make a single cell suspension. After centrifugation, they were resuspended in DMEM medium containing 10% serum and seeded in culture dishes. 2 Cultured in a humidified incubator. The full amount of medium was changed 1 day after inoculation, and the medium was changed every 3 days thereafter. When the cells reached 80% to 90% confluence, 0.25% trypsin was digested and passaged at a ratio of 1:2, and the MSCs were gradually purified by using the differential adhesion property. Cells in the third passage with good growth state were taken, and the monoclonal antibodies CD29 and CD34 were used for flow cytometry to identify...

Abstract

The invention discloses specific application of MSCs (mesenchymal stem cells), solves the problem that no drug for effectively treating the organophosphate-induced delayed neuropathy exists in the prior art, and provides a novel train of thought and specific application for treating the neuropathy. In the application, a chicken suffering from the organophosphate-induced delayed neuropathy is used as an experiment object, and the gait score, the spinal cord solochrome cyanine dyeing detection result, the scanning electron microscope detection result, the P0 protein detection result and the low molecular weight neurofilament detection result are taken as detection indexes; the result shows that the MSCs used as a drug have remarkable recovery effect on chickens suffering from the organophosphate-induced delayed neuropathy, and play an important role in the treatment of occupational poisoning-induced delayed neuropathy.

Description

technical field [0001] The invention relates to the application of a bone marrow mesenchymal stem cell, in particular to the application of a bone marrow mesenchymal stem cell as a medicine for treating organophosphorus-induced delayed neuropathy. Background technique [0002] Organophosphorus-induced nervous system damage in patients with delayed-onset neuropathy is caused by organophosphorus compounds such as tri-o-tolylphosphate, methamidophos, and usually occurs 1-3 weeks after acute poisoning. Its pathological features are axonal swelling and degeneration of distal peripheral nerves and lateral cords of spinal cord, formation of axonal cohesive duct cysts, and secondary demyelination. Many scholars have studied the pathogenesis of organophosphorus-induced delayed-onset neuropathy from the aspects of nervous system cholinesterase inhibition, calcium ion homeostasis imbalance and mitochondrial damage, but so far it has not been very clear. At present, the treatment of pa...
