Abstract

The invention discloses a method for induction of lycium ruthenicum murr tetraploid through tissue culture. The method comprises the following steps that (1) aseptic seedlings are obtained, and blades are picked; and (2) the blades of lycium ruthenicum murr are used as explants of tetraploid induction to be placed on a lycium ruthenicum murr tetraploid callus induction culture medium for callus induction, specifically, the lycium ruthenicum murr tetraploid callus induction culture medium comprises 0.125 mg.L<-1> of 6-BA, 0.025 mg.L<-1> of 2,4-D and 2% of DMSO and 0.01 mg.L<-1> of colchicines. The tetraploid calluses of the lycium ruthenicum murr are successfully induced through combination of dimethyl sulfoxide with the colchicines, and the method lays a foundation for induction of lycium ruthenicum murr tetraploid plants.
