Embodiment 1

[0026] Isolation and identification of Bacillus amyloliquefaciens LM-Y, and its antagonism test on strawberry botrytis cinerea, wheat scab, and peach brown rot. Botrytis cinerea, Fusarium head blight, and Monilia fructigena described in this example are provided by the Laboratory of Plant Pathology, School of Agriculture and Biotechnology, Zhejiang University (conventionally known, Common pathogens that are routinely preserved in various phytopathology laboratories).

[0027] 1. Separation: Collect soil from Fenghua Taoyuan in Ningbo, Zhejiang, weigh 10g of soil sample, add 90ml of sterile water and shake for 2 hours to form a suspension, take 1ml of liquid and add it to a test tube filled with 9ml of sterile water and shake to form 10. -2 Dilution, 10-fold serial dilution to 10 -6 , draw 0.1 mL from each gradient dilution on the beef extract peptone medium, and use a sterile glass coating rod to spread the bacterial solution evenly on the beef extract peptone plate. The sme...

Embodiment 2

[0041] The preparation of embodiment 2 Bacillus amyloliquefaciens LM-Y bacterial agent

[0042] According to the following main steps to complete the preparation of strain LM-Y inoculum:

[0043] (1) The Bacillus amyloliquefaciens LM-Y obtained in Example 1 was inoculated into the beef extract peptone medium, and the activated strain was cultivated at 30° C. for 24 hours; the beef extract peptone medium medium was prepared: beef extract 0.3g, peptone 0.5g, NaCl 0.3g, agar 2g, water 100mL, pH 7.2-7.4, autoclaved at 121°C / 20min.

[0044] (2) inserting the strain activated in step (1) into LB liquid medium, and culturing with shaking at 30° C. and 180 r / min for 24 hours to obtain seed liquid. LB liquid medium preparation: peptone 10g / L, sodium chloride 5g / L, yeast extract 10g / L, distilled water as solvent, pH 7.0, 121℃ / 20min autoclaving.

[0045] (3) The seed liquid obtained in step (2) was inoculated into the beef extract peptone liquid medium at an inoculation amount of 5% by...

Embodiment 3

[0046] Example 3 Verification of field effect of Bacillus amyloliquefaciens LM-Y bacterial agent on peach brown rot

[0047] 3.1 Test drugs

[0048] Bacillus amyloliquefaciens LM-Y bacterial agent (the method of Example 2 prepares the fermentation broth, dilutes it 800 times with water, and the bacterial concentration is 1.0 × 10 7 cfu / mL or so); thiophanate-methyl, tebuconazole (commercially available).

[0049] 3.2 Test crops and control objects

[0050] Test crops: peaches, the variety is Yulu

Abstract

The invention discloses bacillus amyloliquefaciens and application thereof in preparation of antibacterial agents for plant pathogenic fungi. The bacillus amyloliquefaciens LM-Y is separated from rhizosphere soil of diseased peach plants. Compared with the common biocontrol fungi, the bacillus amyloliquefaciens does not produce sensitization spores and is safe to humans, livestock, fruits and vegetables and environmental-friendly. The bacillus amyloliquefaciens LM-Y disclosed by the invention has obvious antagonistic effects on multiple pathogenic fungi such as monilinia fruticola, gibberella zeae, grey mould germs of strawberry and the like, has an inhibition rate of 80% or higher on the monilinia fruticola, has the antibacterial rate higher than that of the conventional biocontrol fungi by about 10% and has great research and development potential. The bacillus amyloliquefaciens LM-Y disclosed by the invention has an obvious effect for controlling the monilinia fruticola, has the field trial control effect of 78.5%, is simple in production process and has very wide application prospects in biocontrol of plant diseases.

Description

[0001] (1) Technical field [0002] The invention relates to a Bacillus amyloliquefaciens and its application, in particular to a Bacillus amyloliquefaciens and its application in the biocontrol of peach brown rot. [0003] (2) Background technology [0004] Peach brown rot is also known as sclerotinia, fruit rot, and fruit rot. The pathogens causing peach brown rot are the subphylum Monilinia fructicola and Monilinia fructicola. fructigena), Monilinia laxa. This disease is a worldwide distribution disease, which can be parasitic on peach, apricot, plum, cherry, plum and other stone fruit trees, causing fruit rot, flower rot and leaf blight. Peach brown rot can cause severe flower rot and leaf blight in the spring flowering and leaf development period if it encounters low temperature and rain; in the later period of growth, if it encounters rainy and humid weather, it can cause fruit rot and lose the economic value of the fruit. At present, the prevention and control of peach...
