The method comprises the following steps: disinfecting Peucedanum praeruptorum explants; then inoculating callus induction mediums with the Peucedanum praeruptorum explants and carrying out callus induction and callus differentiation culture; then selecting cluster buds of the explants and carrying out rooting culture; and finally subjecting tissue culture seedlings to domestication and transplanting and establishing the regeneration system of Peucedanum praeruptorum plants. According to the invention, disinfection effect on the explants is good, and the survival rates of the explants are high; calluses are high in induction rate, great in size and good in growth state; the cluster buds are high in propagation coefficient; the tissue culture seedlings are high in rooting rate and high in survival rate after transplanting, and culture conditions for the tissue culture seedlings are not influenced by insect diseases and changes in the weather; and the method is low in cost, economic, practical, good in repeatability, short in production and growth period and applicable to industrial seedling production of Peucedanum praeruptorum.
