Abstract

The invention discloses a method for measuring the sensitivity and positioning of cholesterol in cells and a diagnostic reagent thereof. The method comprises four steps: (1) separating fresh PBMC cells, carrying out smearing and fixing free cholesterol; (2) carrying out specific dual fluorescent probe incubation of endoplasmic reticulum and mitochondria; (3) performing dual probe labeling; and (4)quantifying and correcting fluorescence in cells. The results show that the content of cholesterol in blood is not in a definite positive relationship with the degree of atherosclerosis and the concentration of cholesterol in tissues of aorta and coronary artery. In a human body, there are multiple cholesterol pools; under the action of a certain pathogenic factor, cholesterol is redistributed with respect to organelle; and thus compared with measurement on the concentration of cholesterol in blood, it is more important to detect and position the cholesterol level in cells.
